Objective:1.Explore the characteristics of dopamine(DA)metabolism in children with attention deficit hyperactivity disorder(ADHD);2.Study the effect of Rhynchophylline(Rh)on the metabolism of DA energy neurons in ADHD model mice;3.Observe the effects of Rhynchophylline(Rh)on the behavior and DA metabolism of dopamine transporter knockout mice.Methods:1.According to the inclusion criteria,we collected and analyzed the clinical data of 89 children with ADHD and healthy children and used ELISA to detect the contents of dopamine metabolism in the two groups of children.2.neurons in vitro ADHD model mice and normal mice,divided into four groups:healthy controls,DAT model group,DAT + Rh,DAT + NDG group,each group after the intervention 0,1,3,6,12 h cells and cell culture collection,and ELISA method to detect the contents of dopamine metabolism in cell cultures of related substances.3.32 DAT knock-out model mice were randomly divided into 4 groups and drug intervention: DAT-KO group、Meth group、NDG group、Rh group.The other eight C57 mice were healthy controls(CTR group).After the intervention,the mice behavior was tested in the 0,7,14 and 21 days,and the contents of dopamine metabolism in the serum of each group were detected.Results:1.The contents of DA,HVA,MAO and BDNF in children with ADHD were significantly reduced(P < 0.01).2.In vitro cultured neurons,the metabolism of dopamine in the DAT model group reached its peak in time 3 and 6h.DA concentration compared with DAT model group and healthy controls,DAT + Rh,DAT + NDG group and moving medium,content of particle group were significantly lower(P < 0.05),the concentration of HVA,MAO concentration,concentration of BDNF were significantly increased,and DAT + Rh DAT + NDG and moving particles there was no significant difference between groups.3.Compared with the blank control mice,the dopamine metabolism decreased significantly in DAT-KO mice(P < 0.01).In the opening experiment,the total activity,average motion speed,time of stay in the central region and the number of standing times increased significantly(P < 0.001);The accuracy of autonomic alternation response in Y maze was significantly reduced(P < 0.001).4.The concentration of DA,HVA concentration,MAO concentration and BDNF concentration were significantly increased in DAT+ Meth group,DAT+ NDG group,DAT+ Rh group(P<0.05).5.DAT + Meth,DAT + NDG group,DAT + Rh group before and after dosing each.The residence time,average speed,the middle area and the number of stand were lower(P < 0.05),but with DAT-K model control group,blank control group,there are significant differences(P < 0.05).Conclusion:1.There is a low level of dopamine metabolism in children with adhd.2.It can promote the metabolism of dopaminergic neurons in vitro.3.It can promote dopamine metabolism in DAT-KO mice.4.DAT-KO mice can be reduced by Rh and Ningdong granule,while improving learning and memory ability.5.The Rh and Ningdong granule can treat attention deficit hyperactivity disorder is related to promoting dopamine metabolism.
