| ObjectiveThe purpose of this study was to explore the expression level of tumor suppressor gene HIC-1 and oncogene HMMR in hyperplasia of mammary gland flocculus,breast fibroadenoma and breast cancer tissue,as well as their relationship with the clinical pathological parameters in breast cancer.Analysis of HIC-1 and HMMR gene expression in breast cancer cell lines MDA-MB-231,as well as HIC-1’s relationship with downstream target gene HMMR.Methods(1)The expression of HIC-1 and HMMR was examined by immunohistochemistry on57 tissue samples which done the operations from ZhouPu Hospital of Shanghai Pudong district(17 cases of hyperplasia of mammary gland flocculus,20 cases of breast fibro adenoma and 20 cases of breast cancer).Through the method of semi-quantitative analysis,the expression between these two genes and the clinicopathological parameters in breast cancer were also analyzed.(2)The SaRNA-HIC-1 respectively transfection breast cancer cell lines MDA-MB-231,after RT-PCR and Real-time PCR method,fluorescence quantitative detection of HIC-1and HMMR gene expression in the cells,and the two genes correlation analysis.Results(1)HIC-1 was moderately positive in samples of hyperplasia of mammary gland flocculus and fibroadenoma(average score 5.65),while the expression of HIC-1 was weakly positive in samples from breast cancer(average score 2.15).The expressing difference of HIC-1 between mammary benign diseases and breast cancer is significant(P <0.05).(2)The expression of HMMR gene in samples of mammary benign diseases was low positive(average score of breast fibroadenoma 0.3,average score of lobular hyperplasia 0),while the expression of HMMR in breast cancer was weekly positive(average score 2.3)(P <0.05).(3)No obvious relationship was observed between the protein expression of HIC-1 and HMMR with clinicopathological parameters in breast cancer.(4)After saRNA-HIC-1 transfection MDA-MB-231 cell lines,HIC-1 mRNA expression increased,at the same time the expresson of HMMR mRNA decreased(P <0.05).HIC-1and HMMR which by the regulation of downstream genes showed negative correlation in breast cancer cells MDA-MB-231.Conclusion(1)The expression of tumor-suppressor gene HIC-1 was significantly lower in breast cancer,while the expression of oncogene HMMR was significantly higher in breast cancer,and it especially expressed in tumor infiltration front area.It indicated that HMMR protein play a role in tumor cells surrounding matrix.(2)HIC-1 and HMMR which by the regulation of downstream genes showed negative correlation in breast cancer cells MDA-MB-231.The findings can provide new molecular targets of breast cancer with certain theoretical and experimental data support.Also the finds have certain clinical value for the molecular mechanisms of development,diagnosis and treatment of breast caner. |
