| Collagen sponge is a kind of sponge prepared by freeze-drying of type I collagen extracted from animals.It is widely used as a medical material for hemostasis and repair of wounds.Currently,medical collagen is mainly extracted from the connective tissue of pigs,cattle and other mammals.However,their safety and economy have been widely questioned due to the transmission of zoonoses,the high price and the limited source of raw materials.Relatively speaking,aquatic collagen represented by fish has the advantages of low cost,extensive source and high biosafety.Therefore,the development of new fish-derived collagen that can replace or supplement the traditional bovine(or pig)collagen has become a research hotspot in many relevant fields at home and abroad.The fish collagen sponge R and S used in this study were independently developed by shandong international biotechnology park.The main raw material was collagen extracted from tilapia skin,which was freeze-dried and then made into a spongy with loose and porous structure.These materials are planned to be developed as a wound hemostatic dressing with the effect of hemostasis and promoting healing.The main content of this study is to evaluate their physical properties,biosafety,and hemostatic properties.1、The tensile strength,microstructure,porosity,water absorption,thermal stability and enzymatic stability of the two sponges were determined.The results showed that the tensile strength of collagen sponges R and S was 0.57±0.13 MPa and 0.11±.0.04 MPa;elongation at break was 6.32% and 31.99%;porosity was 91.25% and 97.38%;water absorption rate was 33.6 times and 19.9 times;thermal denaturation temperature was 49.7± 3.5°C and 46.1 ± 1.6°C;enzymatic hydrolysis time were 85 ± 7.1 min;electron microscopy results showed that both structures are loose and porous,the material R fibers are arranged regularly,the material S is relatively disordered and their structures were more loose.Through experimental tests,it was proved that both sponges have good physical and chemical properties such as water absorption and porosity,and can meet the mechanical performance requirements of fish collagen sponge as a wound dressing.2 、 The biological evaluation of two collagen sponges were peformed by hemolysis test,cytotoxicity test in vitro,and subcutaneous implantation test in rats.The hemolysis and cytotoxicity results proved negative.Collagen sponge R was almost completely degraded at 2 weeks,while collagen sponge S was degraded at 4 weeks,with weak inflammatory reaction and no obvious rejection reaction,proving its good biocompatibility.3、The hemostatic properties of the two materials and the biological activity of repairing wounds were tested by coagulation test in vitro,liver hemostasis test,femoral artery hemostasis test and wound repair test in rats.The results showed that collagen sponge S had a better comprehensive hemostatic effect in different hemostatic tests,slightly better than the listed collagen products,and sponge R had a significantly better hemostatic effect than the negative control group.Both of the two materials had different healing effects on rat skin wounds.The results of physical and chemical index test and biological evaluation showed that both R and S could meet the requirements of mechanical properties of medical devices,and had good effects on hemostasis and repair of wound surface.The hemostatic effect of fish skin collagen sponge R is more prominent. |
