Pseudomonas aeruginosa(PAO1)is a gram-negative bacterium,which is very easy to infect people with low immunity and is a common clinical pathogen at present.In the process of infection,the flagellum is a movement organs of Pseudomonas aeruginosa,which plays an important role in the pathogenesis of PAO1.In previous study,it was found that two peptides which have high homology with two sites(B and P site)of flg E pseudomonas aeruginosa.And the two peptides could bind to corneal epithelial cells.Therefore,in this study,to explore the the effect of flg E gene on virulence factors including molity,biofilm,pyocyanin and type III protein.I respectively constructed the B and P site single mutants(PAO1(flg E-?B),PAO1(flg E-?P))and the double mutants(PAO1(flg E-?BP))in Pseudomonas aeruginosa(PAO1).The results showed that some virulence factors(biofilm,pyogenin,type III secretory protein)significantly increased,the weaken of molity ability in the PAO1(flg E-?BP).The molity ability and the type III secretory protein have no change and biofilm and pyocyanin decreased in the PAO1(flg E-?B).In PAO1(flg E-?P)mutants,expect for the increased of biofilm,the change of other virulence factor is consistent with PAO1(flg E-?B)mutants.The change of these virulence factor is independent on PQS system in these mutants by extraction and analysis of PQS.Above all,B and P site of flg E gene play an important role in the virulence factor,so the flg E gene can as a target in the process of clinical treatment..
