Objective:To discover unknown LncRNAs associated with aneurysmal Subarachnoid Hemorrhage(aSAH)in peripheral blood.Methods:Through the case-control study,75 of the first aSAH patients diagnosed by DSA in hunan region between January 2017 to December 2018 from the Internal Medicine-Neurology,Hunan provincial people’s hospital were collected.A total of 35 cases of normal control group were selected at the same period which had similar information in gender,age,region.5 pairs of clinical aSAH group and control peripheral blood samples were texted in this experiment by high-throughput sequencing methods,GeneSpring GX v12.1software was used to get the expression profiles of LncRNAs,differentially expressed LncRNAs were identified based on fold change and p-value.The differentially expressed LncRNAs were firstly verified in the original high-throughput samples by RT-qPCR to narrow the range,and then the small range differentially expressed LncRNAs were repeatedly verified in the peripheral blood samples of 70 patients with aSAH and30 patients in the control group by expanding the samples.Results:The optimal 482 up-regulated and 543 down-regulated LncRNAs were detected.Expand the sample RT-qPCR method validation ultimately selected two LncRNA: ENST00000505844,ENST00000608142.Area under the ROC curve results of ENST00000505844,ENST00000608142 were 0.806,0.734 respectively.It has certain diagnostic significance.These genes were all up-regulated in aSAH patients obviously and these outcomes were consistent with the sequencing results.Conclusion:1.The differentially expressed LncRNAs were detected preliminarily.2.Extended sample RT-qPCR verification,two up-regulatedLncRNAs,ENST00000505844 and ENST00000608142,were found to be closely related to the occurrence of aSAH. |
