Ultrashort Wave Therapy Of Human Umbilical Cord Mesenchymal Stem Cells On Rabbit Knee Bone Effect Of IL-1β And TNF-α Levels In Synovial Fluid Of Arthritis

Objective To observe the effects of ultrashort wave on the levels of IL-1βand TNF-αin the synovial fluid of rabbit knee osteoarthritis treated with human Umbilical Cord Mesenchymal Stem Cell(hUC-MSCs).Methods 26 healthy New Zealand white rabbits were selected,all of them were 6 months old and male.The right hind limb was made into an experimental model by modified Hulth method.After the model is made,two of them are selected arbitrarily to check whether the model is successful or not.Then the remaining 24 rabbits were divided into four groups by random number table method.The first group was the control group,the second group was the ultrashort wave group,the third group was the stem cell group,and the fourth group was the ultrashort wave+stem cell group.The first group:no intervention after moulding;the second group:15 minutes of micro-calorie ultrashort wave therapy after moulding;the third group:1 x 10~7 umbilical cord mesenchymal stem cells 0.5 ml intra-articular injection after moulding;the fourth group:1 x 10~7 umbilical cord mesenchymal stem cells 0.5 ml intra-articular injection after moulding,and 15minutes of micro-calorie ultrashort wave therapy.The remaining 24 rabbits were executed after 30 days of treatment in each group.The right knee joint fluid was extracted by syringe to observe and analyze whether there were differences in color and quantity compared with other experimental rabbits.The levels of IL-1βand TNF-αwere measured by ELISA.Then the original incision was opened to observe synovium and articular cartilage.The right medial femoral condylar cartilage was harvested and stained with hematoxylin-eosin(HE),and the histological score of the stained cartilage was made with the optimized version of Mankins score.Results 1.The above four methods were used to treat knee osteoarthritis(KOA)in experimental white rabbits.After 30 days,they showed different therapeutic effects.Gross and pathological observation results showed that the treatment effect of the second group,the third group and the fourth group of ultrashort wave+stem cell group on KOA was significantly stronger than that of the first group of control group,and the fourth group was the first and the most ideal group;the treatment degree of the second group and the third group was similar.2.After 30 days of KOA treatment in rabbits,the Mankins score of the right medial femoral condyle cartilage showed that compared with the first group,the results of the fourth group,the third group and the second group were lower(P<0.05).Compared with the ultrashort wave group,the Mankins score of the stem cell group was lower,but the two groups were lower(P<0.05).There was no significant difference between the two groups(P>0.05).Compared with the stem cell group and the ultrashort wave group,the Mankins score of the two groups was lower and had statistical significance(P<0.05).3.Comparing the contents of IL-1βand TNF-αdetected,it can be concluded that after 30 days of treatment,the contents of IL-1βand TNF-αin stem cell group,ultrashort wave group and stem cell+ultrashort wave group were lower than those in control group,and the differences were significant(P<0.05);the contents of IL-1βand TNF-αin stem cell group were only slightly lower than those in ultrashort wave group.The difference was insignificant(P>0.05),but compared with the stem cell group and the ultrashort wave group,the contents of IL-1βand TNF-αin the stem cell+ultrashort wave group were lower,and the difference was statistically significant(P<0.05).Conclusion The improved Hulth modeling method can successfully and efficiently simulate the process and results of human KOA.The modeling method is simple and easy to operate,and can be completed in 7 weeks.Intra-articular injection of human umbilical cord mesenchymal stem cells and ultrashort wave are effective methods for the treatment of knee osteoarthritis.Ultrashort wave has significant synergistic effect on the treatment of knee osteoarthritis,which may be related to the synergistic improvement of the micro-environment of joint injury sites and the regulation of IL-1βand TNF-αcontents in joint fluid.
