Objective : With the aging of diabetes mellitus complicated with neurodegenerative diseases,the problem of progressive cognitive impairment is becoming more and more serious.traditional Chinese medicine(TCM)has obvious advantages in the prevention and treatment of diabetes mellitus and its complications.On the basis of the previous research of the research group,combined with the knowledge of ancient Chinese herbal medicine,based on the baseline equal ratio increase and decrease design method,the core theory of "phlegm-turbidity-toxin" toxic damage to brain collaterals is the core theory of the pathogenesis of diabetic cognitive impairment.Through the establishment of different pathological models of diabetic cognitive dysfunction,the cognitive function system evaluation,glucose and lipid metabolism and insulin resistance,pathomorphology and function of brain tissue were combined,and the comprehensive classification and analysis were carried out.The correlation between component compatibility and pharmacodynamics was quantitatively described,and the compatibility of components of "Steamed coptics chinensis with wine and Grassleaf sweetflag rhizome" against cognitive impairment of diabetes mellitus was discussed from various angles.In order to reveal the scientific connotation of Coptis chinensis-Acorus calamus component anti-diabetic cognitive impairment and guide the rational use of drugs in the clinical specialty of diabetes mellitus and its complications.Methods:By using the method of baseline equal ratio increase and decrease design,the best compatibility of Rhizoma Coptidis and Acorus calamus components against diabetic cognitive impairment was screened out by establishing different pathological models of diabetic cognitive impairment.The high,middle and low dose groups of "Steamed coptics chinensis with wine and Grassleaf sweetflag rhizome" were designed to verify its pharmacodynamic effect.The related biochemical indexes of rat brain tissue were detected respectively,and the preliminary regulatory mechanism of the compatibility of components of "Steamed coptics chinensis with wine and Grassleaf sweetflag rhizome" was discussed.Results:The best compatibility of the components of "Steamed coptics chinensis with wine and Grassleaf sweetflag rhizome" against diabetic cognitive dysfunction was 4,that is,the total alkaloid of Rhizoma Coptidis was 54.7 mg: the volatile oil 4.17 mg of Acorus calamus(based on the original medicinal materials,Wine steaming Coptis chinensis 0.835: Acorus calamus 2.5g)showed a good pharmacodynamic effect on improving abnormal glucose and lipid metabolism and insulin resistance,enhancing learning and memory ability,and improving pathomorphology and function of brain tissue.The mechanism of its compatibility regulation may be related to anti-lipid peroxidation injury,alleviating the response of related inflammatory pathways,promoting acetylcholine synthesis and enhancing cholinergic pathway.Conclusion:To sum up,"Steamed coptics chinensis with wine and Grassleaf sweetflag rhizome" has a good effect on diabetic cognitive dysfunction,and its best compatibility is that the total alkaloid of Rhizoma Coptidis is 54.7 mg: the volatile oil 4.17 mg of Acorus calamus(based on the original medicinal materials,Alcohol steaming Coptis chinensis 0.835: Acorus calamus 2.5g),the preliminary mechanism may be related to anti-lipid peroxidation injury,alleviate the reaction of inflammatory pathway in brain tissue,promote the biosynthesis of acetylcholine,and enhance cholinergic related pathway.The improvement of learning and memory ability and other related cognitive functions.
