| ObjectiveTo analysis the role of HDACs genetic variation and environmental factors in the risk of type 2 diabetes mellitus,as to provide reference for prevention and intervention of type 2 diabetes mellitus.MethodsA community-based,case-control study was conducted to analyze the association between HDACs genetic variants and environmental factors in type 2 diabetes mellitus.Questionnarie was used to collect the information on general demographic characteristics,smoking,alcohol drinking,exercise and the family history of diabetes mellitus through a face-to-face method.Bio-informatics databases,genetic variation prediction tools,and literature were used comprehensively to selected potential functional genetic variants;TaqMan real-time PCR was used to performe genotyping.Unconditional logistic regression were used to examine associations between environmental factors and genetic variation on type 2 diabetes mellitus.We also applied a comprehensive statistical strategy to investigate the role of gene-gene and gene-environment interactions in type 2 diabetes mellitus.Nonparametric test was used to evaluate the effect of HDACs variants on serum ALT,AST and GGT level.The double data entry was made by Epidata 3.02.Data analyses were performed using SAS 9.4.In the demographic characteristics,the differences between the cases and controls were tested using the t test for continuous variables orχ~2 test for categorical variables.Results1.A total of 1006 subjects(503 cases and 503 controls)were included in this study.The analysis showed no exercise(OR=2.053,95%CI=1.212-3.478),BMI≥24.0kg/m~2(OR=1.677,95%CI=1.060-2.652),SBP≥160mmHg and/or DBP≥100mmHg(OR=1.992,95%CI=1.119-3.545)negatively associated with type 2 diabetes mellitus.2.For rs72792338,the TT or TC genotype was associated with a significantly decrease risk of type 2 diabetes mellitus(OR=0.599,95%CI=0.387-0.928).3.rs113045253 and rs3818291 showed negative additive interaction[S=-0.114,AP=0.924,RERI(95%CI)=1.021(0.491-1.551)].But we did not find any significantly multiplicative interaction between gene and behavioral factors.4.Within the group of cases,nonparametric test showed rs11304525and rs3818291 polymorphism was associated with the level of AST,ALT and LDL(P<0.05).ConclusionsNo exercise,BMI≥24.0kg/m~2,SBP≥160mmHg and/or DBP≥100mmHg are risk factors for type 2 diabetes mellitus.For rs72792338,the TT or TC genotype was associated with a significantly decrease risk of type 2 diabetes mellitus.Interaction between rs11304525and rs3818291 contributed significantly to type 2 diabetes mellitus.Besides,these could influence the level of AST,ALT and LDL. |
