| Objective: To observe the effects of Electroacupuncture on learning and memory ability and Reelin signal(Reelin、Dab1、NR2A、NR2B)expression in SAMP8 mice,and to explore the mechanism of electroacupuncture in treating AD from the perspective of Reelin signal,so as to provide experimental basis for clinical treatment of AD by electroacupuncture.Methods: Twenty-four 6-month-old SAMP8 mice were randomly divided into model group and electroacupuncture group,12 mice in each group,and 12 normal aging SAMR1 mice in the same age were selected as control group.In the electroacupuncture group,mice were fixed with self-made net pocket.Baihui(DU20),Dazhui(GV14)and bilateral Shenshu(BL23)were selected for electroacupuncture treatment.After needling,the positive pole of G6805-I electroacupuncture therapeutic apparatus was connected to Dazhui acupoint without manual stimulation.The negative pole was connected to one side of Shenshu(bilateral Shenshu alternately every day).The parameters of electroacupuncture were continuous wave,frequency was 2 Hz,and current intensity was 1.5-2 mA,in order to cause limb microtremor in mice.The electroacupuncture group intervened once a day at the same time,20 minutes each time,8 days as a course of treatment,a total of three courses,each course interval of 2days,a total of 30 days.The learning and memory abilities of SAMP8 mice were evaluated by Morris water maze,the expression of Reelin signal(Reelin,、Dab1、NR2A and NR2B)protein was detected by immunohistochemistry and Western blotting,and the expression of Reelin signal(Reelin、Dab1、NR2A and NR2B)gene was detected by real-time PCR.Results:1.Morris water maze experiment results: Compared with the control group,the average escape latency of the model group was significantly prolonged,the target quadrant dwell time was significantly shortened and the number of times across the platform was significantly reduced,the difference was statistically significant(P<0.01).Compared with the model group,the average escape latency of EA group was shortened,the target quadrant dwell time was significantly prolonged and the number of times across the platform was increased,the difference was statistically significant(P<0.05).2.Immunohistochemical results: Compared with the control group,the average opticaldensities of NR2 A and Dab1 in the model group increased significantly,while the average optical densities of NR2 B and Reelin decreased,the difference was statistically significant(P<0.01).Compared with the model group,the average optical densities of NR2 A and Dab1 in EA group decreased significantly,while the average optical densities of NR2 B and Reelin increased,the difference was statistically significant(P<0.01).3.Immunoblotting results: Compared with the control group,the relative expression of NR2 A and Dab1 protein in the model group increased significantly,while the relative expression of NR2 B and Reelin protein decreased,with statistical significance(P<0.01);compared with the model group,the relative expression of NR2 A and Dab1 protein in the electroacupuncture group decreased,while the relative expression of NR2 B and Reelin protein increased,with statistical significance(P<0.05).4.Real-time PCR results: Compared with the control group,the expression of NR2 A and Dab1 receptor gene in the model group was significantly increased,while the expression of NR2 B and Reelin receptor gene was decreased,with statistical significance(P<0.01);Compared with the model group,the expression of NR2 A and Dab1 receptor gene in the electroacupuncture group was significantly decreased,and the expression of NR2 B and Reelin receptor gene was increased,,with statistical significance(P<0.01).Conclusion:1.Electroacupuncture at Baihui,Dazhui and Shenshu points can improve the learning and memory ability of SAMP8 mice.2.Electroacupuncture at Baihui,Dazhui and Shenshu points may inhibit the expression of NR2 A and Dab1 protein in hippocampus of SAMP8 mice and promote the expression of NR2 B and Reelin proteins. |
