| Objective:The object of the study is to explore effects and mechanism of Huoxue Tongdu decoction on the inflammation after acute spinal cord injury by observing the regulation of Huoxue tongdu decoction on the inflammation mediated by Microglial Activation.Methods:48 SPF class female SD rats were were divided randomly into 3 groups as Sham group、Model group、HXTDD group,each groups had 16 rats.Rats in Sham group were subjected to laminectomy and in the other groups rats were subjected to acute spinal cord injury by modified Allen’s impact device.It’s continuous lavaging for 3 days of pre-operation and postoperation,Huoxue Tongdu Decoction group was gavaged with Huoxue Tongdu Decoction twice a day,while sham-operation group and model group were both gavaged with normal saline twice a day.3 days after intervention,rats were sacrificed to collect spinal cord tissues.Nissl’s staining was applied to observe the morphology and structure of spinal cord and neurons,The expression of ED-1 protein、NLRP3 protein、cleaved-Caspase1(P20)protein were detected by Immunohistochemistry and Western blot,Elisa detected the expression levels of IL-1β、IL-18.Results:1、Morphological observation: The structure of spinal cord of the Sham group is intact,and a clear Nissl body can be seen in the neurons.HXXD group appear lesser degree of Gore focal and Neurons edema when compared with model group.2、The results of experimental expression:By Immunohistochemistry,Compared with Sham group,The expression of ED-1、NLRP3、cleaved-Caspase1(P20)protein in Model group were significantly increased(P < 0.05).The expression of ED-1 、 NLRP3 、cleaved-Caspase1(P20)protein in Model group were higher than that in HXXD group By Immunohistochemistry(P < 0.05).The expression of ED-1 protein 、 NLRP3 protein 、cleaved-Caspase1(P20)protein in Sham group were lower than that in Model groups(P<0.05).the expression of ED-1 protein、NLRP3 protein、cleaved-Caspase1(P20)protein in HXXD group were lower than that in model group by Western blot(P<0.05).compared with Sham group,expressions of IL-1β,IL-18 in model group were significantly higher by Elisa(P<0.05).compared with Sham group,expressions of IL-1β,IL-18 in HXTDD group were significantly lower by Elisa(P<0.05).Conclusions:Huoxue Tongdu decoction can inhibit inflammation and promote rehabilitation and repairment of neuron after acute spinal cord injury with a mechanism that Huoxue Tongdu decocotion can inhibit neuroninflammation mediated by Microglial Activation. |
