Objective To investigate the effects of the serum of compound Lizard Powder on the proliferation,apoptosis,cycle and expression of TGF-β and PI3K/Akt pathway protein in human gastric cancer cell line MGC-803,and to elucidate the effect of this prescription on the gastric cancer cells.The mechanism of action provides a basis for clinical treatment.Methods 160 SPF SD male rats were randomly divided into 8 groups: normal saline group(group A),5-FU group(group B),huachansu group(group C),compound lizard powder 80 group(Group D),compound lizard powder 100 mesh group(group E),80 and 100 mesh mixed low dose group(group F),middle dose group(group G)and high dose group(group H),a total of 20 each group.Each group was given a corresponding intervention,qd,for 1 week.One hour after the last administration,abdominal anesthesia was performed.Under the sterile conditions,the abdominal aorta was taken for blood and centrifuged to prepare serum containing each group.Human gastric cancer MGC-803 cells were intervened for 24 h,MTT assay was used to detect cell proliferation;TUNEL was used to detect apoptosis;flow cytometry was used to detect cell cycle changes;Western-Blot was used to detect gastric cancer cells AKT,p-Akt,TGF-Beta protein expression level.Statistical analysis was performed using SPSS 20.0 For Windows software.Results1.Effects on proliferation and apoptosis of human gastric cancer MGC-803 cells:(1)Compared with normal saline control group(group A),the serum of drug-containing rats ineach treatment group can inhibit the proliferation of human gastric cancer cell MGC-803.Apoptosis was induced,P< 0.05.(2)The AOD value of compound lizard powder was less than5-FU(group B)and cinobufacin(group C)positive control group,P<0.05;the total apoptosis rate of compound lizard powder group was greater than that of group B and C,P< 0.05.(3)Compound lizard scattered 80 mesh +100 mesh mixed low(F group),medium(G group),high(H group)dose group AOD value is less than compound lizard powder 80 mesh group(D group),compound lizard powder 100 mesh group(Group E),P<0.05;the total apoptotic rate of F,G,H groups was greater than that of group D and E,P<0.05.(4)Compared with the F,G and H groups,the effect of H group on human gastric cancer cell MGC-803 was the most significant P<0.05.2.The effect on the cell cycle of human gastric cancer MGC-803:(1)Compared with the saline negative control group(group A),the serum of the drug-containing rats in each treatment group can block the cells in G1 phase,P<0.05.(2)The proportion of G1 phase cells in compound lizard powder group was higher than that in group B and C,P<0.05.(3)The proportion of G1 phase cells in the compound group of lizard scattered 80 mesh + 100 mesh mixed low(F group),middle(G group)and high(H group)was greater than that of group D and E,P<0.05.(4)Compared with the F,G and H groups,the effect of H group on human gastric cancer cell MGC-803 was the most significant P<0.05.3.The effect of TGF-β and PI3K/AKkt pathway protein expression levels:(1)Compared with the saline negative control group(group A),the expression levels of p-Akt and TGF-βprotein decreased in each treatment group,P<0.05.It indicated that the positive control group B,C and each Chinese medicine group promoted the apoptosis of human gastric cancer cell lines;(2)the compound lizard powder group and the 5-FU(group B)and cinobufacin(group C)positive control group were more protein The expression level decreased,P<0.05;(3)compound lizard scattered 80 mesh + 100 mesh mixed low(F group),medium(G group),high(H group)dose group and compound lizard powder 80 mesh group(D group)Thecompound expression level of the compound lizard powder 100 mesh group(group E)decreased,P<0.05;(4)compound lizard powder 80 mesh +100 mesh mixed low(group F),medium(group G),high(group H)In comparison,the levels of TGF-β and p-Akt protein in group H decreased most significantly,P<0.05.There was no significant change in the overall AKT.Conclusion1.The serum of compound lizard powder-containing rats inhibited proliferation and induced apoptosis of MGC-803 cells cultured in vitro.2.Compound lizard powder-containing rat serum can block MGC-803 cells cultured in vitro in G1 phase.3.Compound Lizard Powder can reduce the expression of TGF-β and p-AKT protein,inhibit the canceration by inhibiting TGF-β and PI3K/AKT signaling pathway,and play a role in inhibiting tumor growth.
