Objectives: To establish renal failure model by adenine gavage.To observe the general conditions,renal function,renal pathological changes,renal cell apoptosis and the expression of CHOP and Caspase-12 in renal tissues after treatment with Upbear the clear and downbear the turbid capsule.Methods:72 rats divided into 6 groups randomly: normal control group,model control group,Shenshuaining Capsule control group,upbear the clear and downbear the turbid capsule in low concentration group,middle concentration group,high concentration group.To establish renal failure model by adenine gavage for 21 days.Each treatment group was treated with gavage for 28 days,once a day,during the treatment period,the normal control group and the model control group were given equal volume of purified water.After treatment,observe the general conditions of rats in each group.The renal function was examined by automatic biochemical analyzer.Using HE staining to observe the pathological changes of kidney.Immunohistochemistry way to observe the expression of CHOP and Caspase-12 in renal tissues.The apoptosis renal cells was detected by TUNEL method.Western blot were used to detect the expression levels of CHOP and Caspase-12 in the renal.Results: After modeling,the renal tubular damage of rat kidney tissue and interstitial fibrosis were obvious,and there was a lot of brown crystal deposition in the renal tubule,chronic inflammatory cell infiltration can be seen.The degree of pathological changes in each treatment group was between the blank control group and the model control group.After treatment,the body weight of the rats in each treatment group was higher than that in the model control group(P<0.05),and each treatment group could improve the general condition of rats.After treatment,the BUN and Scr of the treatment group were lower than those of the model group(P<0.01),but still higher than the blank control group(P<0.01).The renal function of low concentration group of upbear the clear and downbear the turbid capsule was lower than that of the Shenshuaining group(P<0.05);The immunohistochemical expression of CHOP and Caspase-12 was mainly distributed in renal tubular epithelial cells,which were mainly cytoplasmic staining,and the basal expression was observed in the blank control group,both immunohistochemistry and Western blot showed that the expression levels of CHOP and Caspase-12 in the treatment group were lower than those in the model group(P<0.01),compared with the Shennanning group,the expression of CHOP in the low concentration group of upbear the clear and downbear the turbid capsule was lower(P<0.05),and there is no statistically different of the expression of Caspase-12 between the two groups(P>0.05).The results of TUNEL showed that the number of apoptosis was decreased in each treatment group compared with the model control group(P<0.05),and it is most obvious in the upbear the clear and downbear the turbid capsule low concentration group.Conclusions: Upbear the clear and downbear the turbid capsule can inhibit the high expression of CHOP and Caspase-12 in renal tissues,inhibit excessive apoptosis of renal tissue cells and protecting renal parenchyma cells,therefore,the purpose of delaying the progression of slow renal failure is achieved.
