| Objective To investigate the relationship between interleukin-6(IL-6),P-selectin gene and rs2200733 SNPs(single nucleotide polymorphisms)in patients with atrial fibrillation in Ningxia.And the association of serum levels that IL-6 and soluble P-selectin in patients with atrial fibrillation in Ningxia.Methods A case-control study was conducted to collect peripheral blood and serum samples from 485 patients in the General Hospital of Ningxia Medical University and the Cardiovascular and Cerebrovascular Hospital of Ningxia Medical University and the physical examination center of Ningxia Medical University General Hospital.Include 222 patients in the AF(atrial fibrillation)group and 263 patients in the NAF(non-atrial fibrillation)group.Specimen DNA was extracted and the single nucleotide polymorphisms of IL-6 and P-selectin gene-related sites were detected by ionization time-of-flight mass spectrometry(MALDI-TOF-MS).ELISA(enzyme-linked immunosorbent assay)was used to detection the human IL-6 and soluble P-selectin serum concentrations.Results 1.Selected IL-6 gene SNPs,only rs1800796 accorded with HWE genetic balance distribution(P=0.921);2.rs1800796 CC/CG/GG genotypes frequency had no statistically significant difference between AF group and NAF group(P>0.05);3.rs1800796 allele C/G was not significantly different between AF group and NAF group(P>0.05);4.Logistic regression analysis found there was no correlation between rs1800796 and the risk of atrial fibrillation(OR=1.138,95% CI: 0.855~1.514,P=0.375),after correct age,gender,hypertension,diabetes,coronary heart disease and body mass index(BMI);5.There was no statistically significant difference in IL-6 serum concentrations between patients with paroxysmal atrial fibrillation,persistent atrial fibrillation and permanent atrial fibrillation(P>0.05).Meanwhile,between patients with atrial fibrillation carrying different genotypes of rs1800796,There was also no statistically significant difference in IL-6 serum concentrations(P=0.705).6.Chi-square test was used to analyze the genotype frequency and allele frequency of P-selectin gene SNPs locus rs1800805,rs1800807,rs1800808.The results showed that genotypes frequency distribution(P=0.553,P=0.422,P=0.909)and alleles frequencie distribution(P=0.663,P=0.255,P=0.672)were no significant difference in the AF group and the NAF group.7.Using logistic regression analysis,after adjusting for age,gender,hypertension,diabetes,coronary heart disease,and BMI,there was no significant association between the single nucleotide polymorphism site rs1800805 and the risk of atrial fibrillation(OR=1.181,95% CI: 0.0.850~1.662,P=0.312);single nucleotide polymorphism locus rs1800807 is not a risk locus of atrial fibrillation(OR=1.197,95% CI: 0.890~1.611,P=0.312);there was no association between the single nucleotide polymorphism locus rs1800808 and the risk of atrial fibrillation(OR=1.178,95% CI: 0.833 to 1.666,P=0.355).8.There was no statistically significant difference in serum concentration of soluble P-selectin between patients with paroxysmal atrial fibrillation,persistent atrial fibrillation and permanent atrial fibrillation(P>0.05);between patients with atrial fibrillation carrying different genotypes of rs1800805/rs1800807/rs1800808,There was also no statistically significant difference in soluble P-selectin serum concentrations(P>0.05).9.Single nucleotide polymorphism locus rs2200733 near PITX2 gene,its genotype frequency and allele frequency were different in AF group and NAF group(χ2=11.483,P=0.003;χ2=11.337,P=0.001).10.After further inclusion of age,gender,hypertension,diabetes,coronary heart disease and BMI variables,CC genotype was used as a reference genotype.Logistic regression analysis revealed that carried rs2200733 TT(OR=2.376,95% CI: 1.391~4.058,P=0.002),CT(OR=1.792,95% CI: 1.164~2.759,P=0.008)people the risk of atrial fibrillation was higher than carrying the CC genotype.Conclusion 1.IL-6,P selectin gene SNPs rs1800796,rs1800805,rs1800807,rs1800808 were not associated with atrial fibrillation risk in Ningxia;2.The rs2200733 was associated with the occurrence of atrial fibrillation in Ningxia.3.Patients with atrial fibrillation in Ningxia the serum concentrations of IL-6 and soluble P-selectin between different atrial fibrillation types and related SNPs genotypes,there were no significant differences. |
