| Objective To help the prevention and treatment of Henoch-Schonlein purpura in children by the study of the HLA-B gene polymorphism on the pathogenesis and prognosis of children with Henoch-Schonlein purpura in Ningxia,and the association between the main type of HLA-B gene and children’s constitution.Methods Children with Henoch-Schonlein purpura who were hospitalized in Department of Pediatrics,General Hospital of Ningxia Medical University from July 2017 to January2018 were studied as case group while the healthy children in the mean time were randomly selected as control group.The specific method is mentioned below.First,collected blood samples from 66 children with case group and 75 from the control group.The crucial point must be mentioned is the general data between the two groups has no significant differences(P<0.05).Then,observe the detection rate of HLA-B15,B35,B40 gene in blood samples of two groups by sequence specific primer PCR method.Finally,find the differences of three genotypes between the two groups and their effects on the pathogenesis and prognosis of Henoch-Schonlein purpura by the statistical analysis.At the same time,through the physical constitution survey scale and the Chinese syndrome type analysis scale to investigate the relationship between HLA-B genotype and children’s body constitution who had the target gene in the above-mentioned samples.Results First,the positive rates of HLA-B15,HLA-B35 and HLA-B40 genotype had a significant differences between two groups with the HLA-B15 genotype was higher in case group than those in the control group(30.30%vs10.67%),the HLA-B35 and B40 were bothhigher in control group than that in the case group(20.00%vs10.61%,30.67%vs15.15%),the OD between each group was 2.13,0.40 and 0.37.Likely,there was a significant differences among the three genotypes in Henoch-Schonlein purpura and Henoch-Schonlein nephritis(P>0.05).Second,the children with HLA-B15 genotype was detected to be of yin deficiency and the HLA-B35 genotype was more manifested as the phlegm-dampness or the damp-heat,while,the children with HLA-B40 genotype was determined as mild physical constitution..Third,the number of patients with yin deficiency of HLA-B15 was found to be the wind-heat wound syndrome,and the patients with damp-heat or phlegm-dampness of HLA-B35 was was found to be the damp-heat syndrome.Conclusion HLA-B15,HLA-B35 and HLA-B40 are related to the incidence of allergic purpura in children in Ningxia.HLA-B15 may increase the risk of disease while HLA-B35 and B40 wound decrease it.In addition,there was a certain correlation between HLA-B genotype and TCM constitution.The physique types of children with susceptible genes were Yin deficiency,phlegm dampness and damp-heat.The children with susceptible constitution wound express wind-heat TCM syndrome or damp-heat blocking syndrome if they got the disease.Forthermore,Henoch-Schonlein purpura nephritis is more likely to occur in children with damp-heat,and most of them wound occur damp-heat blocking syndrome. |
