Effects Of Overexpression Of JNK1 And JNK2 On Proliferation And Apoptosis Of Human Squamous Cell Carcinoma Cell Lines SCL-1 | | Posted on:2020-08-08 | Degree:Master | Type:Thesis | | Country:China | Candidate:H X Liu | Full Text:PDF | | GTID:2404330596483594 | Subject:Dermatology and sexually transmitted diseases | | Abstract/Summary: | PDF Full Text Request | | Objective To investigate the effects of c-Jun amino-terminal kinase 1(JNK1)or c-Jun amino-terminal kinase 2(JNK2)overexpression on proliferation and apoptosis of human squamous cell carcinoma cell line(SCL-1)and its molecular mechanism.Methods The target genes JNK1 and JNK2 were combined with pHBAD-EF1-MCS-3flag-CMV-GFP vector,successful to build a recombinant adenovirus expression vectors of JNK1 and JNK2,transfected the vector into SCL-1 cells,Real-time quantitative PCR(qPCR)and Western Blot method was used to identify the effect of JNK1,JNK2 gene overexpression after optimized the transfection condition;Using CCK-8(cell counting kit-8)and cell clone formation method to measure the proliferative activity and cell colony forming ability of SCL-1 cells;Cell scratch test was used to detect the cell fusion rate;Flow cytometry was used to analysis cell apoptosis;qPCR and Western Blot method was used to detect the mRNA level and protein level expression of JNK1、JNK2 and c-Jun.Results Construction of JNK1/JNK2 overexpressing adenoviral vector successfully;The optimal transfection conditions of JNK1 and JNK2 overexpressing adenoviral vectors was MOI=100 and the transfected 48h;the expression levels of JNK1 and JNK2 in SCL-1 cells were significantly increased after transfection(P<0.05).Compared with the control group the proliferation and anti-apoptotic ability of Ad-JNK1 group had no significant effect(P>0.05)but Ad-JNK2 group cell proliferation and anti-apoptosis was significantly enhanced(P<0.05);Meanwhile,JNK2 overexpressing could increase the phosphorylated protein expression of c-Jun but JNK1 overexpressing could not.Conclusion JNK2 has the function of enhancing the proliferation and anti-apoptotic ability of SCL-1 human cutaneous squamous cell carcinoma cells. | | Keywords/Search Tags: | cutaneous squamous cell carcinoma, SCL-1 cells, JNK1, JNK2, cell proliferation, apoptosis | PDF Full Text Request | Related items |
