| Objective: To establish a model of severe acute pancreatitis(SAP)in rats at different altitudes,observe the pathological morphology of pancreas and lung tissues,and perform pathological scoring.The expression of p38 MAPK was detected by western-blot and RT-PCR,and p38 MAPK was explored.The difference and clinical significance of SAP combined with lung injury in high altitude hypoxia environment.Methods: A total of 320 Wistar male rats were used as the research subjects.The rats were placed in the 400-meter-high group(Xi’an group),2300 meters above sea level(Xining group)and 3300 meters above sea level(Xinghai group)according to random data method.At different altitudes such as the altitude of 4500 m(Guoluo Formation)(80 in each group),each altitude group was divided into F group(sham operation group),P1 h group,P6 h group,P12 h group,P24 h according to random data method.Group(16 in each group).In the sham operation group,only the pancreas was turned over several times after laparotomy,and the artificial SAP model was established by subcutaneous injection of sodium taurocholate in the pancreas of each time group.Pancreas and liver tissues were taken,fixed with 10% formaldehyde solution,stored in liquid nitrogen and frozen at-80℃.The pathological morphology of pancreas and lung tissues were observed under microscope,and pathological scores were obtained.The changes of p38 MAPK protein activity in pancreatic tissue and lung tissue were detected by western-blot method.The expression of p38 MAPK mRNA in pancreas and lung tissues of each group was detected by RT-PCR.Results: At the same altitude,there was no significant difference in pathological score,p38 MAPK protein expression and p38 MAPK mRNA expression between pancreatic tissue and lung tissue in sham operation group(P>0.05).Pancreatic tissue of rats in each time group of SAP and The measured indexes of lung tissue were significantly increased and continued to increase with time,and there were significant differences.At the same time point,the expression of p38 MAPK protein and p38 MAPK mRNA in pancreatic tissue and lung tissue of SAP group were significantly different(P>0.05),and it also showed a continuous increase with the increase of altitude conditions..In the hot spring areas with the highest altitude,the pathological scores of pancreatic tissue and lung tissue,p38 MAPK protein expression and p38 MAPK mRNA expression were the most significant.Conclusion: Under the same altitude condition,the damage degree of SAP lung injury gradually increased with the prolongation of the disease course,and the pathological damage of pancreas and lung tissue and the activation of p38 MAPK protein continued to increase.At the same time point,the damage degree of SAP lung injury increased with altitude.Gradually,the level of p38 MAPK mRNA in pancreas and lung tissue also increased. |
