| Chronic gastritis places a considerable burden on the health care system worldwide.Due to the complex etiology and pathogenesis of chronic gastritis,it is a daunting challenge for chronic gastritis therapy in modern medicine.Traditional Chinese Medicine(TCM)formulas characterized by multi-compounds and multi-targets have been acknowledged with striking effects in the treatment of chronic gastritis in China’s history.Nevertheless,their accurate mechanisms of action are still ambiguous.In this study,we analyze the active compounds,potential targets and related biological pathways of the five herbs contained in Weili tablets,a Traditional Chinese Medicine(TCM)formula,by systems pharmacology approach to detect their molecular mechanism.83 active compounds with good pharmacokinetic are screened out,most of which have been reported with excellent biological activities,and then 183 potential drug targets interacting with those active compounds are identified by algorithm developed by our laboratory.We find that the potential targets of the active compounds are significantly enriched to the chronic gastritis therapeutic targets by analyzing their biological processes through KEGG,DAVID and CTD database.Based on that,we construct the Compound-Target network,Target-Pathway network and "Chronic gastritis" pathway,which further reveal that Weili tablets probably regulate the inflammation and apoptosis caused by chronic gastritis through multiple pathways and multiple modules of mutual synergy to achieve the purpose of treating chronic gastritis.The analysis of the mechanism of the active ingredients in Weili tablets is an another example of the application of systems pharmacology in traditional which promotes the application of traditional Chinese herbs in modern medicine. |
