Research On The Expression Of MicroRNA-125a-5p And Rab25 In Psoriasis Vulgaris

Objective: The aim of this study is to detect the expression of mi R-125a-5p,Rab25 m RNA and it’s protein in skin tissue of normal person and psoriasis patients,further explore the relationship of mi R-125a-5p and Rab25 protein in psoriasis.Methods: Lesional skin of 20 progressive vulgaris psoriasis and normal skin of 16 healthy human controls were collected.The expression levels of mi R-125a-5p and Rab25 m RNA were detected by q RT-PCR.The expression of Rab25 protein was detected by immunohistochemistry.The measurement data was tested for normality and homogeneity of variance,and the data conforming to the normal distribution was performed using LSD-t test,otherwise the data was performed using rank sum test.Results: 1.The results of q RT-PCR showed that the expression of mi R-125a-5p in the progressive psoriasis vulgaris group 1.118(0.592,1.382)was significantly lower than normal control group 2.270(0.729,8.191),and the differences were statistically significant(P<0.05).The value of Rab25 m RNA in the progressive psoriasis vulgaris group 0.920(0.418,1.631)was higher than the normal control group 0.744(0.314,1.707),and there was no difference between the two groups(P>0.05).No association was found between the expression levels in Rab25 m RNA and mi R-125a-5p of psoriaic skin(r=-0.197).2.The results of immunohistochemical indicated that the expression of Rab25 protein were positive both in the stratum spinosum and granulosum of lesions in active stage of psoriasis vulgaris group and the normal control group.The distribution of major Rab25 protein was in the apical region of the cell and the expression level of Rab25 protein in psoriasis vulgaris group(0.262±0.031)was significantly higher than that in normal control group(0.224±0.021),the difference in statistic was significant(P<0.05).The expression level of Rab25 protein had low negative correlation with mi R-125a-5p(r =-0.309).Conclusion: The expression level of mi R-125a-5p was down-regulated in skin lesions of progressive psoriasis vulgaris,while the expression level of Rab25 protein was up-regulated,suggesting that both may be involved in the pathogenesis of psoriasis.
