Objective: Studies have shown that erxian decoction(EXD)can effectively activate the hypothalamic-pituitary-gonadal axis for the treatment of adolescent delay,but the upstream mechanism of its effect on the hypothalamus is not clear.This paper studies its effect on the Leptin/-msh /Kisspeptin pathway to clarify the upstream mechanism of erxian decoction for the treatment of adolescent delay.Methods: 1.The molecular docking of the main components of EXD and the target of Leptin/Α-msh/kisspeptin was studied by using software such as MOE.2.After 3 days of adaptive feeding of 90 3-week-old male SD rats to basic feed,the group was randomly divided into two groups,one set of 10 as the normal control group with basic feed feeding,and the rest were fed with high-fat feed to prepare the obesity animal model.To 6 weeks old to determine the weight of rats,weight than the normal group of average weight 20% was considered a successful model,the obese rats randomly divided into 5 groups,each group of 10.The rats in the experimental group were filled with low middle and high concentration of EXD,the normal control group and the blank control group were filled with gastric saline,and all were executed after 4 weeks,and the material was taken.He staining observed changes in testicular tissue morphology.The serum T,lh,fsh,leptin index of rats was detected by ELISA.WB detects the expression of Kisspeptin,kiss1 R,Leptin R protein content in the hypothalamus tissue.IHC detected positive expression of Leptin and α-MSH in hypothalamic tissue.Results: 1.Molecular docking results: Epimedium has the highest score,the most stable combination with GPCRs,and interacts with multiple residues,and the binding effect is significantly worse after taking off the sugar base.In addition,the BII glycosides in the Fairy,the mother saponins of the Shiretoko and the Huang ester of Huangbai also showed a better binding effect with the Mc3 r,leptin receptor and the GPCRs.2.To the end of the 10 th norfloxacin,the testicular weight and testicular coefficients in the EXD experimental group were significantly higher than those in the model group(p<0.05).Compared with the model group,testosterone(T)and luteinizing hormone(LH)were increased in the EXD experimental group,and the follicle stimulating hormone(FSH)and leptin(Leptin)decreased,among which the Lh,fsh,leptin difference was statistically significant(p<0.05).He staining showed that the small tubes of testicular tissue in the EXD experimental group and the normal control group and the positive drug group had normal growth,and the spermatogenic cells were closely arranged.In the model group,some spermatogenic tubular walls were thin,the number of spermatogenic cells decreased obviously,the arrangement was more messy,and the occasional shedding cells.Western Blot results showed an increase in the expression of Kisspeptin and Kiss1 R in EXD Experimental group,in which the expression of Kiss1 R was statistically significant(p<0.05),and exd R expression in leptin experimental group and model group decreased compared with normal group.However,no significant differences were seen.Immunohistochemical results showed a decrease in the positive expression of α-MSH in hypothalamic tissue of rats in the EXD experimental Group(p<0.05).Conclusion: The main components of EXD and Leptin R and other stable combination,and then through the α-MSH effect,to a certain extent,so that Kisspeptin expression increase,so as to effectively start the HPG axis,promote the normal reproductive development of puberty in rats.
