Objective:Congenital heart disease is the most common congenital malformation of newborn babies.TBX1 is a key regulator in heart development,and our previous study found that it may be involved in the occurrence of congenital heart disease by affecting TGFβ2.miRNA can inhibit or degrade the translation process by specifically binding to the target messenger RNA and carrying out gene regulation at the post-transcriptional level,which is closely related to a variety of heart diseases.In this study,the expression changes of miR-193a-3p,miR-141-3p and miR-200a-3p were detected after targeted interference of TBX1,to explore the possible pathway for the regulation of TGFβ2 by the TBX1 gene.Method:1.The TBX1-siRNA or Negative control RNA were transfected into H9C2 rat cardiomyocytes respectively,and were divided into TBX1-siRNA group and NC group according to different treatment factors.The protein expression level of TBX1 in H9C2 cells after transfection for 48 h was detected by Western Blot.miR-193a-3p,miR-141-3p and miR-200a-3p were selected for study.2.H9C2 cells transfected with TBX1-siRNA or Negative control RNA for 48 h were collected,and the expression levels of miR-193a-3p,miR-141-3p,and miR-200a-3p were detected by RT-PCR.3.SPSS 21.0software was used to analyze the experimental data,the mean standard deviation was used to represent the measurement data,and t test was used for the comparison between the two groups,and P<0.05 was used to define whether the difference was statistically significant.Result: 1.Western Blot was used to detect the protein expression level of TBX1 in H9C2 cell line 48 h after transfection,and the results showed that the TBX1-siRNA group was significantly lower than the NC group in the protein expression level of TBX1,with statistically significant differences(P<0.05).2.The results of RT-PCR showed that after transfection for 48 h,the expressions of miR-193a-3p,miR-141-3p and miR-200a-3p in the TBX1-siRNA group were significantly down-regulated compared with the NC group,and the differences were statistically significant(P < 0.05).Conclusion:Targeted interference of TBX1 gene expression may down-regulate the expression of miR-193a-3p,miR-200a-3p and miR-141-3p in H9C2 rat cardiomyocytes.
