Objective:Ischemic stroke is a common and serious disease in neurology,with increasing incidence and poor prognosis year by year,it has very high risk disability rate and fatality rate.Hypertension,diabetes,hyperlipidemia and smoking are the main risk factors of cerebral infarction.With the development of research on genetic factors,genetic factors have been found to play an important role in the pathogenesis of ischemic stroke in addition to traditional risk factors.Therefore,the genetic susceptible gene of cerebral infarction can be screened out,and individualized prevention and treatment can be conducted for the susceptible population,so as to reduce the incidence,disability and mortality of cerebral infarction.The purpose of this study was to investigate the correlation between matrix metalloproteinase 12(MMP-12)gene rs660599,low density lipoprotein receptor related protein 6(LRP6)genes rs11054731,rs2302685 and atherosclerotic cerebral infarction(ACI)in Han population of Liaoning area.Methods:A case-control study was conducted.The case group included 213 cases of ACI patients of han nationality in liaoning area,and the control group included 197cases of physical examination during the same period.The ratio of male to female in the case group was 140:73,with an average age of(64.71±10.61)years.In the control group,the ratio of male to female was 112:85,and the age was(62.97±8.99)years.c~2test was used to analyze the purpose gene genotype and allele frequency distribution of the differences between the two groups.The target genes polymorphisms were detected by improved multiplex ligation detection reaction(IMLDR),SPSS24.0statistical software was adopted to analysis statistic,Logistic regression model was used for multivariate analysis to exclude the influence of traditional risk factors.P<0.05 showed statistical difference.Results:1.The results of the Hardy-Weinberg law of genetic balance test on each polymorphism locus of MMP-12 and LRP6 genes in the case group and the control group showed that the genotype and allele frequency of the population in this study were consistent with the genetic balance(P>0.05),and the population was well representative and suitable for genetic analysis.2.There were no significant differences in age,gender,triglycerides,cholesterol and apolipoprotein A1 levels between the two groups(P>0.05).The common risk factors of stroke,such as hypertension,diabetes,smoking,drinking history and low density lipoprotein cholesterol,were significantly higher in the case group,with statistically significant difference(P<0.05).3.There was no statistical difference in MMP-12 gene rs660599 genotype distribution and allele frequency between the case group and the control group(P>0.05).4.The GG genotype and G allele at rs11054731 of LRP6 gene in the case group were significantly higher than those in the control group(P<0.05).Further model construction and analysis showed that the recessive model had statistical significance between the two groups(P<0.05).After adjusting for traditional risk factors with conditional logistic regression,the G allele frequency of LRP6 gene rs11054731 was significantly higher than that of the control group,and the difference was statistically significant(P<0.05;OR=1.46).5.There was no statistical difference in the genotype distribution and allele frequency of LRP6 gene rs2302685 between the case group and the control group(P>0.05).Conclusion:1.The G allele of LRP6 gene rs11054731 may be correlated with the susceptibility to atherosclerotic cerebral infarction in the han population in liaoning area.2.The polymorphism of MMP-12 gene rs660599 may have no significant correlation with cerebral atherosclerotic cerebral infarction in the han population in liaoning area.3.The polymorphism of LRP6 gene rs2302685 may have no significant correlation with cerebral atherosclerotic cerebral infarction in the han population in liaoning area.
