Objective:There is a novel mitochondrial open reading frame of the 12S rRNA-c derived from the mitochondrial open reading frame,which is expressed in various tissues of humans and animals,and mainly acts on blood circulation.Skeletal muscle,which increases glucose uptake and utilization,thereby improving insulin resistance and regulating metabolic balance.This study mainly revealed whether aerobic exercise and energy control activate AMPK/PGC-1α pathway,increase MOTS-c expression,and improve the signaling mechanism of insulin resistance.Methods:Animal models were established,energy control,exercise,and drug intervention were given,and animal models were randomly divided into 12 groups.After the insulin resistance model was established,the high-fat group was still fed with high-fat diet,and the energy-controlled group controlled the diet to 70-%and replaced it with normal feed.At the same time,exercise intervention and drug intervention were given.The exercise intervention was treadmill exercise:lh/day,every Friday,the drug intervention was continuous intraperitoneal injection of MOTS-c for 70 days.Mouse intra-peritoneal glucose tolerance test(IPGTT)and ELISA measured MOTS-c concentration,Western blot was used to detect signal pathway expression.Results:1.The expression of MOTS-c in serum and skeletal muscle increased significantly after exercise,and the aerobic exercise promoted the expression of MOTS-c.2.Compared with the normal control group,the expression level of MOTS-c was significantly lower in the high-fat diet group.After aerobic exercise,drug intervention and joint intervention,the expression level of MOTS-c was significantly increased.3.The expression level of MOTS-c in the energy control group was also significantly higher than that in the high-fat diet group.4.AMPK/PGC-1α pathway pair The expression of MOTS-c has a regulatory effect,and the effect of MOTS-c is similar to that of aerobic exercise.Conclusion:1.AMPK/PGC-1α pathway cascade kinase is involved in mediating mitochondrial MOTS-c expression,and PGC-la acts as an upstream regulator of MOTS-c expression;2.Aerobic exercise activates AMPK/PGC-1α pathway,Increase the expression of MOTS-c and improve the mechanism of insulin resistance;3.Dietary control increases the utilization of sugar by skeletal muscle,while increasing the expression of MOTS-c and improving insulin resistance.
