Objective:To investigate the the effect of long-term high-intensity interval exercise on the inflammatory cytokine of skeletal muscle with different muscle fibers.Methods:18 young SD male rats aged 8 weeks were randomly divided into the quiet group(group N),the medium-high intensity endurance exercise group(group M),and the high-intensity interval exercise group(group H).The rats of group N do not make any intervention,and exercise group were trained on treadmill with the slope of 0 °using tail electrical stimulation and manual driving.Fitness training: group M was to train at a speed of 15m/min for 20 minutes for 6 days,resting on Sunday.Group H was to train at a speed of 25m/min for 2 minutes and 15m/min for 2 minutes,6groups a day for 6 days.Formal training: group M was to train at a speed of 20m/min for 30 minutes at the first week,training 25m/min for 30 minutes at the second week,training 30m/min for 30 minutes at the third week,training 35m/min for 30 minutes at the 4-8 week.Group H was to train at a speed of 30m/min for 2 minutes and15m/min for 2 minutes at the first week,training 35m/min for 2 minutes and20m/min for 2 minutes at the second week,training 40m/min for 2 minutes and20m/min for 2 minutes at the 3-8 week.Group M was trained once a day and group H was trained 6 groups a day,and they were both trained for 6 days,resting on Sunday.The rats fasting 24 hours after training were taken by intraperitoneal injection of 20%Ulla sugar(5ml/kg)anesthesia,and the soleus and extensor digitorum longus muscle of rat legs were quickly separated and removed.The gene and protein expression of IL-6,TNF-α,P65 and RelB in soleus and extensor digitorum longus muscle were detected by fluorescence quantitative PCR and western blot.All data were processed by SPSS16.0 software.Results:1.In soleus and extensor digitorum longus muscle,the expression of IL-6mRNA in group H was significantly higher than that in group M(P < 0.05),while the gene expression of TNF-α,P65 and RelB in each group showed no significant difference(P > 0.05).2.The protein expression of IL-6 in the cytoplasm of soleus and extensor digitalis longus muscle was significantly higher in group H than in group N and M(P < 0.05),and there was no significant difference in TNF-α,P65,P-P65,RelB and P-RelB protein expression between groups(P > 0.05).3.There was no significant difference in the protein expressions of P65 and RelB in soleus and extensor digitorum longus muscle nucleus between groups(P > 0.05).Conclusion: 1.Long-term HIIT can up-regulate gene and protein expression of IL-6in the soleus muscle and extensor digitalis longus muscle of normal rats.2.Long-term HIIT did not affect the classical and non-classical NF-κ B signaling pathways in the soleus and extensor digitorum longus muscle of normal rats.
