Cyclocarya paliurus(Batal.)Iljinskaja is a cyclocarya plant.In recent years,reports on the cyclocarya mainly focus on its hypoglycemic activity.And diabetes,as a chronic lifelong disease,has seriously threatened human health.Therefore,the screening of Hypoglycemic Active fractions,preparation technology,quality standard and fingerprint of polysaccharide fractions of cyclocarya leaves will be studied in this paper,which will provide a basis for the comprehensive utilization of Cyclocarya.In this study,the model of hyperglycemia induced by alloxan in mice was used to evaluate the effect of different extracts of cyclocarya on blood glucose of alloxan-induced hyperglycemia mice with 0.5,1.0 and 2.5 h blood glucose values.The results showed that 70% ethanol precipitation of cyclocarya(QQL-70)significantly reduced blood glucose levels compared with the model group(P < 0.001).With the PNPG method,and the ?-glucosidase inhibitory activity as an indicator.To investigate the inhibition rate of ?-glucosidase in ethanol precipitation at different concentrations of cyclocarya.The results showed that when the ethanol concentration was 70%,the ?-glucosidase inhibition rate was good,reaching 96.71%.The model of hyperglycemia induced by alloxan in mice was used to investigate the effect of QQL-70 on fasting blood glucose in mice with hyperglycemia induced by alloxan.The area under the blood glucose curve was used as an indicator to investigate the four pairs of QQL-70.The effect of oxypyrimidine on glucose tolerance in hyperglycemic mice,the results showed that compared with the model group,the positive drug group and the QQL-70 dose group had no effect on fasting blood glucose,and the positive drug group and the high dose group had significant effects on the glucose tolerance of alloxan-induced hyperglycemia mice(P< 0.01).Experiments showed that QQL-70 had the effect of increasing glucose tolerance,and it can be determined that the ethanol concentration of the ethanol precipitation process was 70%.In this paper,the fingerprint of cyclocarya leaves was established by high performance liquid chromatography,methodological investigation,similarity analysis,cluster analysis and principal component analysis were carried out to investigate the best origin of medicinal materials.The results showed that 8 common peaks were calibrated,and the relative retention time and relative peak area RSD of each common peak were less than 5.0%,which proved that the method is simple and reproducible,and can be used for the control of medicinal materials quality.The results of cluster analysis and principal component analysis showed that the quality of the medicinal materials in Jiangxi Xiushui was the best,which was consistent with the real estate area in Jiangxi Province.The water extraction process and alcohol extraction process were investigated by orthogonal test method.The extraction rate and the proposed amount of polysaccharide were used as indicators to inspect the water extraction process.The best water extraction process is to add 25 times amount of water,boil 3 times for 1 hours each at the temperature of 100?.After water extraction,70% ethanol precipitation was used to complete the extraction of medicinal materials.Taking hygroscopicity,solubility,forming rate and fluidity etc as indexes,the best type of excipients and ratio were examined,and taking moldability as the index,the best type ofoptimal amount of excipients and wetting agents(Ethanol)concentration was examined;taking mouth feel as an indicator,the optimum amount of flavoring agent was examined.Take the dry paste powder obtained by the above extraction process,according to 1:5 add Mannitol-lactose-salose(1:2:1)auxiliary material and 1.0% mogroside and mix well,use 80% ethanol as wetting agent,granulation.The colorimetric method was used to establish a method for determining the content of polysaccharides in the preparation.Referring to the relevant general rules of the 2015 edition of the Chinese Pharmacopoeia,the inspection items related to the difference of particle size,moisture,solubility and loading under the granules were carried out,and the results met the requirements.
