| Mycomedicine Tremella sanguinea,also known as blood fungus,is called the blood mushroom because the pigment of the fruiting body turns red after it dissolves in water.It belongs to Eumycota,Basidiomycetes,Tremellales,Treimellaceae,Tremella.Traditional medicine records that T.sanguinea is benefiting qi and activating blood circulation,clearing heat and dampness,nourishing yin and fluid.This paper is the firs research to systematically study the chemical constituents of fruiting body of the T.sanguinea in order to screen some compounds that inhibit tumor cells and antibacterial activity,to provide a theoretical basis for the rational development and utilization of fungi and edible fungi resources.This article first evaluated the authenticity of the Tremella sanguinea,by observing its traits,microscopic characteristics and physical and chemical phenomena,the fungus was identified as T.sanguinea.In chemical composition research,the chemical constituents of the fruiting body of T.sanguinea were systematically separated and purified by silica gel,Sephadex LH-20 and RP-HPLC.Their structures were identified by physicochemical properties,spectroscopic analysis,as well as comparisons with the data reported in literature.33 compounds were isolated from the 90%ethanol extract of the fruiting bodies of T.sanguineaear fungus,among which,27were identified and 6 were not identified.They were identified as(22E)-5α,8α-epidioxy-24-methyl-cholesta-6,9(11),22-trien-3β-ol(1),(22E)-5α,8α-epidioxyergosta-6,22-dien-3β-ol(2),cerevisterol(3),ergosta-7-ene-3β,5α,6β-triol(4),(22E)-6β-methoxyergosta-7,22-diene-3β,5α-diol(5),ergosta-7-en-3β-ol(6),4-hydroxy-17R-methylincisterol(7),uridine(8),2-pyrrolidone(9),nicotinamide(10),1-(3-indolyl)-3-dihydroxypropan-1-one(11),yangambin(12),2-hydroxybenzoic acid(13),2,3-dihydroxybenzoic acid(14),4-hydroxybenzoic acid(15),(9Z,12Z,15Z)-octadeca-9,12,15-trienoic acid(16),Linoleic acid(17),(9Z,12Z,15Z)-2,3-dihydroxypropyl octadeca-trienoate(18),2,3-dihydroxypropyl 9Z,12Z-octadecadienoate(19),glycerol monooleate(20),crypticin B(21),dibutyl phthalate(22),bis(2-ethylhexyl)phthalate(23),terephthalic acid bis(2-ethyl-hexyl)ester(24),3-phenyllactic acid(25),3-methoxybenzoic acid(26),Benzoic acid(27).Among them,the compounds 1 to 7 were steroidal compounds,the compounds 8 to 11 were alkaloid compounds,the compound 12 was a lignan compound,the compounds 13 to 15 were phenolic compounds,and the compounds 16 to 20 were fatty acid compounds.All compounds were isolated from T.sanguinea for the first time,except for compounds6,9,15,17,and 22,the other compounds were isolated from the genus Tremella for the first time.Compounds 28 to 33 were to be further determined for their structural formula.Using the GC-MS analysis method,11 compounds were identified from the fruiting body of Tremella sanguinea,and 8 compounds were identified from the petroleum ether extract of Tremella sanguinea.In biological activity research,27 monomeric compounds were screened for antibacterial activity by microdilution method.The tested strains were Micrococcus tetragenus,Bacillus cereus,Bacillus subtilis,Staphylococcus aureus,Staphylococcus albus,and Escherichia coli.The results showed that compound 7 showed significant inhibitory activity against Micrococcus tetragenus,Staphylococcus aureus,Staphylococcus albus and Escherichia coli.The MIC for S.aureus,S.albus and E.coli was 2.5 ug/mL,and the MIC for M.tetragenus was 10 ug/mL.Except for this,the other compounds showed no significant antibacterial activity,and the MIC were all greater than 10μg/mL.Twenty-two monomeric compounds were tested for cytotoxic activity using MTT method,the tested tumor cell lines were Hela,H460,and B16F10.The results showed that all tested samples had no significant cytotoxic activity,the IC500 were all greater than 40μmol/L. |
