| ObjectiveAsthma is a chronic airway inflammatory disease which seriously threatens the physical and mental health of children.Up to days,pathogenesis of asthma have not entirely been explored.The imbalances of Th1/Th2 and Th17/Treg in the process of child asthma are regarded as one of important pathogenesis.In this study,the percentage of T-cells subgroups,Treg and Th17 in the peripheral blood of children with asthma was detected by flow cytometry,and the expression of Th1,Th2,Th17 and Treg cytokines in the serum of children with asthma during the acute phase was detected by flow cytometry CBA,as well as the effect of Tim-3 acting on the imbalance of Th1/Th2 and Th17/Treg in the serum of children with asthma,so as to explore the imbalance of Th1/Th2 and Treg/Th17 in children with asthma.Methods1.81 children with asthma(asthma group)and 38 children who were hospitalized for minor operation such as hydrocele or hernia without any symptoms of asthma(control group)were recruited in this study,who were admitted to the Qilu Children’ Hospital of Shandong University from October 2013 to December 2014.All asthmatic children were diagnosed asthma or stop inhaled hormone therapy more than 3 months after the recurrence.2.Collection of two groups of children with peripheral blood,the levels of T-cells subgroups,Treg and Th17 were determined by flow cytometry.3.The expression of serum Th1,Th2,Treg and Th17 cytokines IL-2,IL-6,IL-10,TNF,IFN-γ,IL-17 A and IL-12/IL-23p40 was measured using CBA method.4.Additionally the level of Tim-3 mRNA in the serum of children with asthma was detected by RT-PCR.Results1.There were no differences in age,sex between the asthma group and control group;2.Compared with the control group,each age group the proportion of cells in CD4 T,CD3T and less than 2 years old CD4/CD8 T were significantly lower(P<0.01 or P<0.05),the proportion of CD8 T cells in various age groups and more than 2 year old CD4/CD8 T in the two groups had no significant difference(P>0.05);3.Compared with the control group,the percentage of Th17 in the peripheral blood of children with asthma was increased(P<0.05),and the percentage of Treg in the peripheral blood of the two groups had no significant difference(P>0.05);4.Compared with the control group,serum Th1 cytokines IL-2 and IFN-γ in children with asthma were significantly increased(P<0.05),and the content of TNF in serum of the two groups had no significant difference(P>0.05);5.There was statistically significant that asthma group serum Th2 cytokine IL-4,IL-6 were lower than the control group(P<0.01 or P<0.05);while the IL-10 concentration was higher than control group(P<0.05);6.The asthma patients serum Treg cytokine IL-12/IL-23p40 was significantly lower than control(P<0.01),and there was no significant difference between the two groups in the expression of Th17 cytokine IL-17A;7.Compared with the control group,the relative expression of serum Tim-3 mRNA in asthma was markedly high(P<0.05).Conclusion:With Th1 and Th17 cells hyperfunction,the function of Th2 and Treg cells decreased,and Th1/Th2 and Treg/Th17 cytokine imbalance may play an important role in the pathogenesis of asthma with children.And Tim-3 may have an effect on the imbalance of Th1/Th2 and Treg/Th17 in children asthma. |
