The occurrence and development of breast cancer are closely related to dietary factors, especially dietary patterns.Current research on the effects of different dietary approaches on tumor progression has focused on carcinoma in situ and metastatic cancer, but there is no effective way to monitor the process of tumor metastasis in vivo.In this article, mice were subjected to dietary intervention from childhood to youth.Mice tumor models were constructed at the age of eight weeks in mice. The effects of dietary patterns on tumor metastasis in mice were evaluated by monitoring the number of circulating tumor cells(CTCs)during tumor metastasis.Dietary therapy is a promising strategy to prevent drug resistance and reduce side effects in cancer treatment that can provide more valuable advice for people to dietary properly.Based on the in vivo capture method of CTCs,this paper combined the size and weight of carcinoma in situ and the number of lung metastatic cancers to evaluate the effects of dietary patterns on breast cancer metastasis. The method based on in vivo capture of CTCs has a positive effect on early detection of cancer, and this article is to simultaneously evaluate the effects of multiple dietary patterns and dietary patterns combined with food functional factors on breast cancer metastasis. The paper mainly consisted of four parts,as follows:In chapter one,the CTCs and their capture and detection methods were summarized, and the research progress of the effects of different dietary patterns and food function factors on cancer was introduced.CTCs and their conventional detection methods are briefly outlined, and the research progress of the effects of different dietary patterns and the three food functional factors such as hesperetin, allicin, and astragalus polysaccharide on cancer were highlighted. In chapter two,the effects of different dietary patterns on breast cancer metastasis were evaluated based on CTCs in vivo capture methods. The experiment began with dietary intervention in mice at an early age,and 4T1 xenograft models were established at the age of eight weeks in mice. The number of CTCs was used to reflect the status of cancer metastasis. The increase in the number of CTCs was significantly promoted by dietary patterns such as High-salt diet,High-sugar diet and High-fat diet, while delayed by Ketogenic diet,Low-fat diet,Low-protein diet,Dietary restriction and Mediterranean diet. These results indicated that the in vivo capture and detection of CTCs provides a convenient method for real time monitoring of cancer metastasis and spread.Different dietary patterns profoundly affect the metastatic process of cancer, and have profound meanings in improving the effect of cancer treatment and reducing drug side effects. In chapter three, the functional indwelling needles were used to evaluate the effects of the combined effects of the Mediterranean diet and food functional factors such as hesperetin, allicin, and astragalus polysaccharide on breast cancer metastasis based on CTCs in vivo capture methods. One of the dietary pattern with the best anti-cancer metastatic effect screened in Chapter 2: Mediterranean diet, combined with anti-cancer food functional factors such as hesperetin, allicin and astragalus polysaccharides, which have obvious anti-cancer effects,to intervene in the dietary of mice. The effects of the Mediterranean diet combined with food functional factors on the number of CTCs in mouse tumor models were investigated and compared with individual dietary pattern interventions.Meanwhile, combined with the volume and weight of carcinoma in situ and the number of lung metastatic cancers, the effects of the Mediterranean diet combined with food functional factors on breast cancer metastasis were comprehensively evaluated. These results indicated that the combined effects of Mediterranean diet and food functional factors on cancer metastasis were not as good as that of a single Mediterranean diet, which showed that some ingredients in food may have an antagonistic effect, which provides some suggestions for people to adjust their dietary structure and reasonable dietary. In chapter four, the effects of different dietary patterns on breast cancer metastasis based on the in vivo capture method of CTCs were summarized, and the effects of Mediterranean diet combined with food functional factors on breast cancer metastasis were analyzed.According to the number of CTCs,the cancer metastasis process can be monitored in real time during the cancer metastasis process. The selected healthy dietary patterns have a positive effect on improving the treatment of cancer, preventing drug resistance and reducing side effects, and provide valuable suggestions for people to have a reasonable dietary.
