| Objective:The chronic asthmatic rat model was established by ovalbumin combined with aluminum hydroxide sensitization and ovalbumin atomization.Yanghe Pingchuan Granules was used to intervene the chronic asthmatic rats,observe its effect on airway inflammation,and preliminarily explore the effect of Yanghe Pingchuan Granules on Notch signaling pathway.Based on the previous research,the rat model of chronic asthma was established by sensitization of ovalbumin combined with aluminum hydroxide and atomization challenge of ovalbumin.Yang Hechuan granule was used to interfere with chronic asthma rats,and its effect on airway inflammation and Notch signal pathway and its mechanism were observed and discussed.Methods:Forty male clean SD rats were randomly divided into 4 groups by random number table method,which were normal group,model group,Yanghe Pingchuan Granules group and positive drug group,with 10 rats in each group.After adaptive feeding,ovalbumin combined with multiple injections of aluminum hydroxide were used to sensitize and stimulate rats in the other groups except the normal group to establish a chronic asthma rat model,and then the Yanghe Pingchuan Granules group was given Yanghe Pingchuan Granules solution(7.74 g/ kg),the positive drug group was given dexamethasone solution(0.125 mg/kg)once a day for 6 weeks;observe the general changes of the rats during the modeling,record the weight and death.After the intervention,samples were taken,and pathological changes such as inflammatory cell infiltration and smooth muscle changes in lung tissue sections were observed and evaluated using HE staining.Ig E,IL-4,and IFN-γ levels in serum were measured by ELISA;Mouse Notch1,GATA-3 and T-bet protein expression in lung tissue;RT-q PCR method was used to detect Notch1,GATA-3 and T-bet m RNA expression in lung tissue.Results:1.The general state of rats in each group: in the experiment,the breathing,reaction,hair,nose scratching and diet of rats in the normal group were normal,while in the model group,the response was slow,restless,the hair was disgraceful and fluffy,the food intake was slightly reduced,and the respiratory frequency was more rapid.Frequent nose scratching,cough,audible wheezing and wheezing were also found in the positive drug group,and the performance of the positive drug group was better than that of the model group.Compared with the model group,the initial improvement of the Yang Heping Chuan Granules group was not obvious,but the symptoms in the later stage were better than that of the model group.Three rats in the positive drug group died in the course of the experiment,while there was no death in the rest group.There was no significant difference in pre-model weight and pre-administration weight among the three groups(P>0.05).Compared with the model group,the weight gain of the positive drug group was less than that of the model group,and there was significant difference between the two groups(P<0.01).There was no significant difference in pre-death weight among the other groups(P>0.05).2.Pathological changes of lung tissue in each group: in the normal group,the airway structure of lung tissue was intact,the bronchial lumen was smooth,there was no obvious infiltration of inflammatory cells in airway and surrounding vascular tissue,the morphology and structure of smooth muscle was normal,and there was no glandular hyperplasia.There was no alveolar fusion or septal thickening.The results of lung histopathology in the Yanghe Pingchuan Granules group and the positive drug group were lighter than those in the model group,and the lung tissue structure was more complete,with a small amount of inflammatory cell infiltration around,less bronchial contracture,no thickening of smooth muscle,and bronchial lumen close to normal.3.Compared with the model group,the levels of Ig E and IL-4 in the Yanghe Pingchuan Granules group decreased significantly(P<0.01),the levels of Ig E and IL-4 in the positive drug group decreased significantly(P<0.05),and the levels of IFN-γ in the positive drug group increased significantly(P<0.01).4.Compared with the normal group,the expression of Notch1,GATA-3 protein and m RNA in the model group was significantly increased(P<0.01),and the expression of T-bet protein and m RNA was significantly decreased(P<0.01);compared with the model group,the expression of Notch1,GATA-3 protein and m RNA in the Yanghe Pingchuan Granules group was significantly decreased(P<0.01),and the expression of T-bet protein and m RNA was up-regulated(P<0.05);Compared with the model group,the expression of Notch 1,GATA-3 protein and m RNA in the lung tissue of the positive drug group also decreased(P<0.05),and the expression of T-bet protein and m RNA increased(P<0.05).Conclusion:1.Yanghe Pingchuan Granules can reduce the pathological changes of lung tissue and airway inflammation in rats with chronic asthma.2.Yanghe Pingchuan Granules can reduce the levels of IL-4 and improve the levels of IFN-γ in serum of chronic asthmatic rats,and then intervene the airway inflammation of chronic asthmatic rats.3.Yanghe Pingchuan Granules can regulate the expression of Notch signal pathway related protein and gene level in lung tissue of chronic asthma model rats.4.The intervention of Yanghe Pingchuan Granules on airway inflammation and inflammatory factors IL-4 and IFN-γ in rats with chronic asthma may be through the regulation of Notch signaling pathway. |
