AimTo explore the differential proteins expression in filum terminale of children with tethered cord syndrome(TCS)and normal fetus by label free quantitative and analyze the pathogenesis of TCS.MethodThis study was a randomized controlled study,which have experimental group(EG)and control group(CG).We randomly selected 3 cases of filum teriminale samples from children with TCS as EG,who were diagnosed and treated by surgical operation in our hospital from November 2018 to June 2019.Another 3 cases of normal filum terminale without nervous system malformation induced by other reasons were CG.All the patients’ family were informed of the experimental purpose and signed the informed consent form.All the samples were taken from near the end of the spinal conus for protein extraction.The differential proteins of the two groups were screened by label free quantification combined with liquid chromatography tandem mass spectrometry technique,and the bioinformatics analysis of differential proteins were carried out by using MaxQuant database and Uniport database.ResultAfter the identification of the two sample groups,253 differentially expressed proteins were obtained,including 101 up-regulated proteins and 152 down-regulated proteins.The differential proteins were analyzed by bioinformatics analysis,such as GO(Gene Ontology)enrichment analysis,KEGG(Kyoto Encyclopedia of Genes and Genomes)enrichment analysis and protein-protein interaction analysis.The results indicated that proteins with large differences,such as apolipoprotein Al(APOA1)and complement C3(C3)are involved in the process of lipid metabolism,whileα-enolase(ENO1)and biglycan(BGN)are involved in the process of fiber degeneration.SDEPs were involved in the processes of lipid metabolism,fiber degeneration,coagulation cascade,ribosome transformation,and carbon metabolism.ConclusionThere is a difference between the proteins of EG and CG.The pathway and metabolic pathways which the proteins involved in may play an important role in pathogenesis of the filum terminale disease and participate in the occurrence of TCS.
