Background and ObjectiveWith the advancement of science and technology and the development of society,wireless network has penetrated into everyone's life.Wifi is popular because of its fast,convenient and free acess to the internet,but its impact on human body and other organisms has not been a clear research result.Chian has established grading standards for electromagnetic radiation,first level standards(safety standards,no impact on human body),and second level standards(middle area,harmful impact on human body).It has been reported abroad that electromagnetic radiation has adverse effects on organisms even within the specified safety range.Scientists in the Netherlands have found that wifi radiation may be the main cause of tree wilt.The researchers also found that wifi radiation slowed the growth of corn cobs.However,Tsinghua University has done on seed germination undrer the radiation of wifi radiation.The research results found that wifi radiation has no harm to plant growth and does not affect seed germination.Moreover,according to existing research and relevant international standards,wifi radiation has no significant impact on human health.At present,scholars at home and aroad have just begun to study this kind of situation,but there is no report at home and abroad on the influence of electromagnetic radiation on some specific biological activities,such as the repair of nerve injury and its mechanism.In this study,we will study whether the repair of sciatic nerve injury is affected by wifi radiation in rats,and if so,we will further study its mechanism and prevention methods.So as to lay a foundation for the next study whether wifi affects the repair of human nerve injury and how to reduce or eliminate it.Methods72 male SD rats were randomly divided into experimental group,control group and sham operation group,24 rats in each group.Take intraperitoneal anesthesia at a dose of 1%sodium pentobarbital(5mg/kg).The rats were fixed on a rat rack,routinely disinfected,spread a towel,and took a posterolateral incision of the right lower limb,about 6 to 8mm in length.The biceps femoris and lateral femoris muscles were exposed,and the sciatic nerve was separated by blunt separation.The experimental group and the control group in 7mm away from the lower edge of the priformis,the sciatic nerve was clamped 3 times,10 seconds each time and 10 seconds in the middle.After washing with normal saline,the incision was sutured layer by layer.The experimental group rats were palced near the router,the control group and the sham-operation group were placed in the Faraday cage,no electromagnetic radiation was given,and the cage was protected with tin foil to prevent other electromagnetic radiation interference.The operation steps of the sham operation group was the same as that of the experimental group and the control group.Only the sciatic nerve was isolated and not clamped.The sciatic nerve function index(SFI)and BBB-score of rats were measured and immunohistochemistry was performed to measure the expression of brain-derived neurotrophic factor(BDNF)?nerve growth factor(NGF)and growth associated protein-43(GAP-43)in the sciatic nerve at 4?8 and 12 weeks after operation,the pathological changes of the sciatic nerve were observed under a light microscope.Results(1)Behavioural tests:The three groups of the sciatic nerve function index at the 4rd weeks were 0?—88.652±0.541?—89.725±0.613 respectively,at the 8rd weeks were 0?-79.313±0.782?-80.432±0.733 respectively,and at the 12th weeks were 0?—77.293±0.742?—78.654±0.695.The three groups of the BBB scale at the 4rd weeks were 21?8.13±2.04?7.73±2.12 respectively,at the 8rd weeks were 21?9.73±1.86?9.64±2.04 respectively,and at the 12th weeks were 21?11.32±2.21?11.42±1.86.The SFI and BBB scale of the control group and the experimental group were significantly lower than that of the sham operation group at 4?8 and 12 weeks after operation,the difference was statistically significant(p<0.05).There was no significant difference between the control group and the experimental group at 4?8 and 12 weeks after operation(p>0.05).(2)HE staining:At 4?8 and 12 weeks after the operation,the results of HE staining showed that the sciatic nerve adventitia of the sham operation group was complete,the nerve fibers were arranged orderly,the structure was tight,the myelin sheath and axon structure were complete,and a small number of Schwann cells could be seen.In the control group,the sciatic nerve fibers were arranged disorderly,with Wallerian degeneration accompanied by myelin sheath breakdown,a large number of vacuoles,and Schwann cell proliferation was obvious.The sciatic nerve fibers of the experimental group were disordered,with Wallerian degeneration and myelin sheath breakdown.There was no significant difference between the experimental group and the control group.(3)Immunohistochemical staining:The expression of BDNF?NGF and GAP-43 in the sham operation group?the experimental group and the control group were analyzed semi-quantitatively at 4?8 and 12 weeks after operation.The expression of BDNF was 0.143±0.021?0.703±0.051 and 0.654±0.032 respectively in the sham operation group?the experimental group and the control group at 4 weeks after the operation;The expression of BDNF was 0.151±0.018?0.682±0.043 and 0.631±0.029 respectively in the sham operation group?the experimental group and the control group at 8 weeks after the operation;The expression of BDNF was 0.138±0.023?0.605±0.042 and 0.562±0.021 respectively in the sham operation group?the experimental group and the control group at 12 weeks after the operation,with statistical difference between the three groups(p<0.05).The expression of NGF was 0.123±0.023?0.422±0.042 and 0.408±0.035 respectively in the sham operation group?the experimental group and the control group at 4 weeks after the operation;The expression of NGF was 0.109±0.018?0.401±0.035 and 0.391±0.036 respectively in the sham operation group?the experimental group and the control group at 8 weeks after the operation;The expression of NGF was 0.112±0.019?0.385±0.040 and 0.371±0.034 respectively in the sham operation group?the experimental group and the control group at 12 weeks after the operation,with statistical difference between the three groups(p<0.05).The expression of GAP-43 was 0.125±0.031?0.323±0.032 and 0.305±0.023 respectively in the sham operation group?the experimental group and the control group at 4 weeks after the operation;The expression of GAP-43 was 0.112±0.028?0.301±0.029 and 0.287±0.021 respectively in the sham operation group?the experimental group and the control group at 8 weeks after the operation;The expression of GAP-43 was 0.120±0.030?0.283±0.033 and 0.261±0.018 respectively in the sham operation group?the experimental group and the control group at 12 weeks after the operation,with statistical difference between the three groups(P<0.05).The expression level of BDNF?NGF and GAP-43 in the experimental group was similar to that in the control group at 4?8 and 12 weeks after operation,with no statistical difference between the two groups(p>0.05).ConclusionWifi radiation has no significant effect on the repair of sciatic nerve injury in rats.Wifi radiation has no significant effect in the SFI?BBB score?HE staining and the expression of BDNF?NGF and GAP-43 between the experimental group and the control group.
