Research purposes:In order to study the effect of Hugan Yifei Recipe on attenuating(preventing and preventing adverse reactions such as anti-tuberculosis drug liver damage)and to preliminary explore the material basis,the subject first started with the water and alcohol extracts of Hugan Yifei Recipe and the split Hugan Recipe,analyze the scientificity and rationality of the"attenuating effect" of prescriptions from a macro perspective;then find out the existing material components from Hugan Fang,and then screen out the potential effective ingredients as the material basis of Hugan Fang to verify;finally,compare the monomer composition with Hugan Fang to analyze the efficacy,and study the treatment of liver damage Mechanism.Research methods:1.Evaluate the model by light microscope observation of liver HE staining and serum biochemical indicators,and study the water extracts of Hugan Yifei Recipe and Hugan Recipe from physiological,pathological,and behavioral indicators Attenuating effects of alcohol and alcohol extracts on drug-induced liver injury.Automatic biochemical analyzer detects serum alanine aminotransferase(ALT),aspartate aminotransferase(AST),total bilirubin(TBIL),alkaline phosphatase(ALP)and other biochemical indicators;ELISA test The kit detects liver tissue SOD activity and MDA and GSH content determination;RT-PCR method is used to detect the expression of CYP2E1 mRNA in mouse hepatocytes,and qPCR method is used to detect the expression of Mrp2 mRNA in liver cells to investigate the signal pathways it plays a regulatory role Determine its mechanism of action.2.UHPLC-Q-TOF/MS technology was used to detect the components of the five flavors of Hugan Fang to infer the true basic ingredients of the effective substances.Consult the domestic and foreign literature to establish a component library,use Peakview 1.2 software to identify chemical components in the sample,and combine with MarkerView 1.2.1 software to conduct principal component analysis on the sample.Research results:1.The combination of isoniazid and rifampicin can cause drug-induced liver injury for two weeks,which proves the success of modeling.Hugan Yifeiquan Recipe and Hugan Recipe groups can significantly inhibit the increase of total bilirubin(TBIL)(P<0.01)and alkaline phosphatase(ALP)in the serum of liver injury model mice(P<0.05).),Also can inhibit the increase of malondialdehyde(MDA)in liver tissue(P<0.01),enhance the activity of superoxide dismutase(SOD)and the level of glutathione(GSH)in liver tissue(P<0.05)).Observation of pathological sections also showed that liver decoction of Hugan Yifei Quanfang and Hugan Decoction were significantly reduced compared with the model group.2.The obtained data was imported into Peak View 1.2 software.The chemical composition with a matching degree of more than 85%was identified and confirmed.A total of 156 compounds were identified under the positive and negative ion modes.3.For the combination of the five ingredients,the TBIL and ALP were significantly smaller than the model group,and the differences were statistically significant(P<0.05).The glycyrrhizic acid,polydatin,and peony root and other components were not good for attenuating effects.There was no significant difference in ALP(P>0.05).Conclusion:1.Hugan Yifei Recipe and Hugan Recipe can significantly inhibit the increase of TBIL and ALP,significantly increase the levels of SOD and GSH in liver homogenate of drug-induced liver damage mice and inhibit the increase of MDA in liver tissue;pathology Light microscopy observation after HE staining showed that it could reduce the degree of degeneration and even necrosis of liver tissue in drug-induced liver damage mice,and had the effect of repairing liver cells and restoring liver function.The mechanism of action was related to the antioxidant effect.2.The active ingredients in Hugan Recipe have been preliminary explored to inhibit TBIL and ALP in serum,increase SOD and GSH and decrease MDA,indicating that oleanolic acid+schisandrol A+glycyrrhizic acid+polygoside glycoside+eugenin This kind of monomer composition has protective effect on liver injury in model mice,but it does not reach the general attenuating effect,but it shows that its material basis is effective-Significance of the project:By studying the mechanism of attenuating the effects of Hugan Yifei Recipe and preliminary exploring its material basis,it will provide theoretical basis for the clinical use of Hugan Yifei Recipe in the clinical treatment of tuberculosis,and develop Chinese medicine prescriptions or traditional Chinese medicine preparations for the adjuvant treatment of tuberculosis Help a lot.
