Effective Material Basis Research Of Qianyang Yuyin Granules For Renal Protection Of Hypertension

Objective:To analyze the chemical constituents of Qianyang Yuyin Granules systematically,and to study the active ingredients and mechanisms of Qianyang Yuyin Granules in preventing and treating hypertensive renal damage.Methods:The LC/Q-TOF-MS technology and Peakview data processing platform were established to analyze the chemical composition of the water exract of Qianyang Yuyin Recipe rapidly and build a mass spectrometry database.Based on this,the ingredients from Qianyang Yuyin Granules may be presented.In addition,the network pharmacology research was used to select the main active ingredients,targets and mechanisms of Qianyang Yuyin Granules in preventing and treating hypertensive renal damage,and the molecular docking was used to test the binding energy of the key components and targets.Results:1.The LC/Q-TOF-MS technology was used to analyze the chemical constituents of the water exract of Qianyang Yuyin Recipe,and a mass spectrometry database was constructed.A total of 133 compounds have been identified,including flavonoid,iridoid,phenolic,triterpenoid,phenylpropanoid,anthraquinone,stilbene,sterone and so on,and the proposed fragmentation pathways of different structural types of compounds have bene summarized.Furthermore,41 compounds were from Bidens pilosa L.,30 from Polygonum multiflorum Thunb.,44 from Cornus officinalis Sieb.Et Zucc.,39 from Scrophularia ningpoensis Hemsl.,30 from Cvathula officinalis Kuan.,and 43 from Alisma orientale(Sam.)Juzep.A total of 99 compounds have been identified from Qianyang Yuyin Granules,of which the structure type is similar to the compounds in the aqueous extract.2.As results of network pharmacology,a total of 44 components and 48 key targets were identified from Qianyang Yuyin Granules against hypertensive kidney damage.These targets were mainly enriched in 252 significant related biological processes such as angiogenesis,response to hypoxia,nitric oxide biosynthetic process and inflammatory response.And 141 significant signaling pathways were involved,such as MAPK,PI3K-Akt,TGF-β,Wnt and Jak-STAT signaling pathway.The results of molecular docking verify the reliability of network pharmacology.Conclusion:The potential effective ingredients of Qianyang Yuyin Granules were clarified,and mechanism of multiple ingredients,multiple targets and multiple signaling pathways of its prevention and treatment of hypertensive kidney damage was revealed,which provide a scientific basis for further research and clinical application of Qianyang Yuyin Granules.
