Objective:To predict the pathway mechanism of intervention of type 2 diabetes mellitus in Zuo Gui Pill Group,after removing prescription and tonifyin Drug Compatibility Group and tonifyang Drug Compatibility Group by network pharmacology,and then to establish a model of type 2 diabetes mellitus mice to verify its predictive pathway mechanism,so as to reveal the mechanism of action of Zuogui Pill and after removing prescription,tonifyin Drug Compatibility and tonifyang Drug Compatibility intervening in type 2 diabetes mellitus.Methods:In the Network pharmacology part:Chemistry Database,TCMSP,Batman,Bidd database,oral bioavailability and drug similarity or credibility was uesd to obtained and screening the active components.TCMSP,drugbank,Batman,UniProt database,Gene Carads and The Comparative Toxicogemics disease gene database was obtained and screening the intervention of type 2 diabetes potential target gene.Constructing of Zuo-Gui Pill,Yin tonic drugs and yang tonic drugs intervention type II diabetes active substance-potential target gene network by the cytoscape.DAVID was database used to analyze target gene enrichment,Obtain candidate genes and functions involved in related pathways.In the animal experiment part:8 week old 60 MKR mice,10 were used as the blank group,and the remaining 50 were injected intraperitoneally with 40mg/kg STZ buffer,After the modeling was completed,randomly divided into model group,Zuo-Gui pill(Decoction),Yin tonic drugs group,Yang tonic drugs,metformin group,another 10 C57B/L6 mice were used as the normal group,and fasting blood glucose was tested every two weeks.After the drug was administered to the stomach for 35 days,and the serum was centrifuged to detect the fasting blood glucose,TC,TG,HDC-L,and LDC-L contents.The kidney tissues of the mice were extracted and tested for western blot to detect pi3k and akt pathway protein expressions level.Results:1.Through network pharmacology prediction,the potential targets of Zuo Gui Pill group,tonifyin drug compatibility group and tonifyang drug compatibility Group can act on the pi3k/akt signaling pathway and intervene type 2 diabetes by promoting the protein expression of related pathway.2.after drug intervention,compared with the model group,the FBG of Zuo Gui Pill,tonifyin Drug compatibility Group,tonifyang drug compatibility Group and Metformin Group decreased significantly(P<0.05).The effect of Zuogui Pill on reducing blood sugar was better than that of tonifyin drugs and tonifyang drugs(P<0.05).Compared with metformin,the hypoglycemic effect between Zuogui Pill and metformin was equivalent(P>0.05).3.TCM group could decreased serum TC?TG content and increase HDL-C content to alleviate the disorder of blood lipid metabolism in mice with type 2 diabetes mellitus in MKR.4.compared with the model group,the expression of pi3k,akt protein was significantly up-regulated in Zuo Gui Pill,tonifyin Drug compatibility,tonifyang Drug compatibility and metformin(P<0.05).Conclusions:1.According to the prediction of network pharmacology,it is found that the compatibility of Zuogui Pill and tonifyin drugs and tonifyin drugs may interfere with the occurrence and development of type 2 diabetes mellitus through pi3k/akt signal pathway.2.combination of Zuogui pills and tonifyin drugs and tonifyang drugs can significantly reduce the blood sugar FBG type MKR 2 diabetic mice,And the effect of Zuogui Pill group was better than that of tonifyin drugs and tonifyang drugs.3.The compatibility of Zuogui Pill and tonifyin drugs and tonifyang drugs can reduce the content of serum TC,TG and increase the content of HDL-C to alleviate the disorder of blood lipid metabolism in type MKR2 diabetic mice.4.combination of Zuogui Pill and tonifyin after removing prescription and tonifyang Drug can intervene in 2 diabetes mellitus by up-regulating the expression of pi3k,akt protein in mice with type MKR 2 diabetes mellitus,and the effect of Zuogui Pill and and the curative effect of Zuogui pills is obviously higher than that of tonifyin drugs and tonifyang drugs.
