Objective: To detect the expression of Annexin A1(Annexin A1),S100A9,Vimentin(Vimentin)and their mRNA in normal gastric mucosa tissue,adjacent tissue,gastric cancer(GC)tissue and GC lymphatic metastasis tissue,analyze the relationship between the expression level and the clinicopathological characteristics of GC,and preliminary explore the role of Annexin A1,S100A9,Vimentin in the occurrence,development,invasion and metastasis of GC and the correlation between the expression levels of the three Provide experimental basis for diagnosis,treatment and prognosis judgment.Methods: 1.Immunohistochemistry(IHC)and in situ hybridization(ISH)were used to detect Annexin A1,S100A9,Vimentin protein and mRNA in normal gastric mucosa tissue,adjacent tissue,GC tissue and GC lymph node metastasis tissue;2.The chi-square test was used to analyze the differences in the expression of Annexin A1,S100A9,Vimentin protein and their’s mRNA between different tissues;the Kappa consistency test was used to compare the IHC and ISH of Annexin A1,S100A9,Vimentin Results;Using rank correlation rank sum test method,analyze the correlation between Annexin A1,S100A9,Vimentin and the relationships with clinical pathological characteristics of GC patients.Through the above methods,the effects of Annexin A1,S100A9 and Vimentin in the occurrence,development and invasion and metastasis of gastric cancer were preliminarily discussed.Results: 1.The positive expression rates of Annexin A1 in 112 normal gastric mucosa tissues,84 paracancerous tissues,112 GC tissues,and 79 GC lymph node metastasis tissues were: 98.21%,71.43%,17.86%,11.39%;The positive expression rates of its mRNA were: 95.54%,79.76%,21.42%,13.92%;the expression of Annexin A1 protein and mRNA in GC was consistent(Kappa = 0.406,P <0.05).The positive expression rate of Annexin A1 and its mRNA in GC and GC lymph node metastasis tissue is significantly lower than that in normal gastric mucosa tissue and adjacent tissues.Annexin A1 is in the pathological evolution of normal gastric mucosa-para-cancer-GC-GC lymph node metastasis Expression is declining.The spearman correlation analysis showed that the expression level of Annexin A1 was positively correlated with the degree of tumor differentiation(r = 0.371,P <0.05).The higher the degree of gastric cancer differentiation,the higher the expression level of Annexin A1;The number was negatively correlated with the TNM stage(r =-0.545,r =-0.231,r =-0.281,P <0.05),the deeper the invasion of gastric cancer,the more lymph node metastases,the higher the TNM stage,the expression of Annexin A1 The lower the level;the patient’s age,gender,and tumor size have nothing to do with the expression level of Annexin A1.2.In 112 normal gastric mucosa tissues,84 paracancerous tissues,112 GC tissues and 79 GC lymph node metastasis tissues,the positive expression rates of S100A9 were: 29.46%,39.28%,68.75%,78.48%;The positive expression of S100A9 mRNA rates were: 25.89%,35.71%,66.96%,74.68%;the expression of S100A9 protein and mRNA in GC was consistent(Kappa = 0.705,P <0.05).In normal gastric mucosa-paracancer-GC-GC lymph node metastasis,the expression showed a gradual upward trend(P <0.05).Correlation analysis showed that the expression level of S100A9 was negatively correlated with the degree of tumor differentiation(r =-0.339,P <0.05).The higher the expression level of S100A9,the lower the degree of gastric cancer differentiation;It was positively correlated with TNM stage(r = 0.339,r = 0.309,r = 0.350,P <0.05).The deeper the invasion of gastric cancer,the greater the number of lymph node metastases,the higher the TNM stage,the higher the expression level of S100A9;The age,sex,and size of the cancer have nothing to do with the expression level of S100A9.3.In 112 normal gastric mucosa tissues,84 paracancerous tissues,112 GC tissues and 79 GC lymph node metastasis tissues,the positive expression rates of Vimentin were: 0,0,20.53%,29.11%;The positive expression rates of mRNA were: 0,0,17.85%,26.58%;the expression of Vimentin protein and its mRNA in GC were consistent(Kappa = 0.646,P <0.05).Vimentin and Vimentin mRNA were negatively expressed in normal gastric mucosa epithelial tissues and adjacent tissues,and only expressed in the interstitium;partially expressed in GC and GC lymph node metastasis tissues.Correlation analysis showed that the expression level of Vimentin was negatively correlated with the degree of differentiation of gastric cancer(r =-0.310,P = 0.001).The lower the degree of differentiation of gastric cancer,the higher the expression level of Vimentin;and the depth of invasion of gastric cancer,the number of lymph node metastases Was positively correlated with TNM stage(r = 0.223,r = 0.304,r = 0.222,P <0.05),the deeper the invasion of gastric cancer,the more lymph node metastases,the higher the TNM stage,the higher the level of Vimentin expression;and patient’s age,gender,and cancer size were irrelevant.4.Correlation statistical analysis showed that the expression level of Annexin A1,S100A9 and Vimentin in GC was not correlated with that of Vimentin(r =-0.016,P = 0.863);the expression level of both Annexin A1 and S100A9 was negatively correlated with that of S100A9(r =-0.355,P = 0.000);the expression level of S100A9 and Vimentin was positively correlated(r = 0.307,P = 0.001).Conclusion: The expression levels of Annexin A1,S100A9 and vimentin may be closely related to the occurrence,development,invasion and metastasis of GC.
