| [Objective]This work intends to explore whether Hydrogen sulfide(H2S)improve neurogenesis in the hippocampal of Streptozotocin(STZ)-induced diabetic rats,and whether this fuction involves activation of Akt.[Methods]1.Rats were intraperitoneally injected with STZ(55mg/kg),and fasting blood glucose was more than 16.7mmol/L,which was considered as a successful diabetic model.2.SD rats were randomly divided into 5 groups:control group,STZ treatment group,STZ+low-concentration H2S(30 mol/kg)treatment group,STZ+high-concentration H2S(100 mol/kg)treatment group,and single high-concentration H2S(100 mol/kg)treatment group.Diabetic rats were given intraperitoneal injection of NaHS(30μmol/kg)and NaHS(100μmol/kg)once a day for 4 weeks.After the rats were killed,the hippocampal tissue was taken to detect the indicators.3.The expressions of Neurogenesis-associated proteins,including Doublecortin(DCX),Glial fibrillary acidic protein(GFAP),Neuron-specific nuclei protein(NeuN);Akt signaling protein kinase B(AKT)and 473-serinephosphorylated kinase(p-AKT)in hippocampus were detected by western blot.[Results]1.H2S improved hippocampal neurogenesis in STZ-induced diabetic ratsNeurogenesis-associated proteins DCX and NeuN decreased significantly and GFAP increased significantly in STZ-induced diabetic rats.After treatment with Sodium hydrosulfide(NaHS,a donor of H2S,100μmol/kg,i.p.),the expressions of DCX、NeuN protein were increased significantly,the expression of GFAP protein was decreased significantly.These results showed that H2S improved neurogenesis in the hippocampus of STZ-induced diabetic rats.2.H2S activates Akt signaling in the hippocampus of STZ-induced diabetic ratsThe expression of AKT protein had no obvious effect and p-AKT protein decreased significantly in hippocampal of in STZ-induced diabetic rats.After NaHS treatment,there was no significant change in the expression of AKT,while the expression of p-AKT increased significantly.These results showed that H2S improves neurogenesis in the hippocampus of STZ-induced diabetic rats involves the activation of Akt.[Conclusion]H2S can improve neurogenesis in the hippocampus of STZ-induced diabetic rat,which is involved in the activation of Akt... |
