| Objective The natural deep eutectic solvent(NaDES)was used as a green solvent to extract the active ingredients and study the catalytic conversion of the active ingredients in the traditional Chinese medicine Salvia miltiorrhiza.1)Extraction experiment:Green and efficient ultrasonic-assisted extraction of bioactive components from salvia miltiorrhiza by NaDES.Separately,Salvianolic acid B is represented as a hydrophilic component,and tanshinone IIA and cryptotanshinone are represented as a hydrophobic component.2)Catalytic conversion experiment:NaDES is used as a solvent and catalyst to degrade Salvianolic acid B that is rich in Salvia miltiorrhiza into other active ingredients with lower contents in Salvia miltiorrhiza including Salvianolic acid A,Lithospermic acid,Danshensu,Protocatechualdehyde,etc.,laying the foundation for further pharmacological research of the above-mentioned trace active ingredients.In summary,this paper uses natural deep eutectic solvents in the field of green chemistry for the extraction and preparation of active ingredients of traditional Chinese medicine Salvia miltiorrhiza,which improves the utilization of from Salvia miltiorrhiza in extraction and preparation comprehensively.Methods 1.Extraction experiment:22 kinds of natural deep eutectic solvents are used for the extraction of Chinese medicine Salvia miltiorrhiza,and the extraction factors such as material ratio,extraction temperature,extraction time and NaDES content on are optimizated.HPLC is used to detect the experimental results.2.Catalytic conversion experiment:Salvianolic acid B was added to 15 kinds of natural deep eutectic solvents,and the response parameter such as experimental temperature,experimental time,and NaDES content on the degradation reaction of Salvianolic acid B were investigated.HPLC,LC-MS and NMR were used for the qualitative and quantitative analysis of the products.Results 1.Extraction experiment:Compared with water,natural deep eutectic solvent like L-Pro-Lac can significantly increase the extraction of the salvianolic acid B(increased by 42%compared to water)and tanshinone IIA,Cryptotanshinone(Very low tanshinones in water extraction).2.Catalytic conversion experiment:Natural deep eutectic solvents,like Bet-Lac and Bet-Ur,can easily and efficiently convert Salvianolic acid B into Salvianolic acid A,Lithospermic acid,Danshensu,and Protocatechuic aldehyde respectively.Conclusion Using natural deep eutectic solvent as a solvent not only increase the extraction rate of active ingredients of Salvia miltiorrhiza,but also convert Salvianolic acid B into other trace but high efficient Salvianolic acids quickly and efficiently via the catalytic function of natural deep eutectic solvent. |
