| Objective:Based on the analysis of the significantly difference of expression of COX-1 gene in familial aggregation of cerebral thrombosis,sporadic cerebral thrombosis and normal control group,explore the relationship between COX-1 and familial aggregation of cerebral thrombosis.Based on the analysis of the significantly difference of expression of COX-1 protein in familial aggregation of cerebral thrombosis,sporadic cerebral thrombosis and normal control group,explore the relationship between COX-1 protein and familial aggregation of cerebral thrombosis.Methods:Collected 25 cases of familial aggregation of cerebral thrombosis,25 cases of sporadic cerebral thrombosis and 25 cases of normal control group.Using real-time quantitative PCR technique,analyze the significantly difference of expression of COX-1 gene in three groups.Using ELISA method,analyze the significantly difference of protein of COX-1 in three groups.Using statistical analysis,compare whether there is statistical significance(P<0.05)of the difference of gene expression and protein expression in three groups.Results:1.Compared with the group of normal control group,the group of familial aggregation of cerebral thrombosis and sporadic cerebral thrombosis,there was no obvious statistical difference(P>0.05)in the age,history of diabetes,history of smoking,history of drinking,systolic blood pressure,diastolic blood pressure,blood glucose and blood lipid.2.The expression of COX-1 gene in the group of familial aggregation of cerebral hemorrhage and sporadic cerebral thrombosis was obviously decreased compared with the normal control group,and the difference was statistical significance(P<0.05).3.Compared with the sporadic cerebral thrombosis,the expression of COX-1 gene in the group of familial aggregation of cerebral thrombosis was obviously up-regulated,and the difference was statistically significant.4.Compared with the group of normal control group,the group of familial aggregation of cerebral thrombosis and sporadic cerebral thrombosis,there was no obvious statistical difference(P>0.05)in the protein expression of COX-1.Conclusion:The COX-1 gene may be related to the familial aggregation of cerebral thrombosis.The change of COX-1gene expression may increase the susceptibility of cerebral thrombosis.The expression of COX-1 protein may not be related to familial aggregation of cerebral thrombosis. |
