| Objective: To study the regulatory effect of qigui blood-enriching granules on immunosuppression induced by cyclophosphamide in mice and its possible mechanism.Methods: The model of immunodeficiency mice was constructed by intraperitoneal injection of cyclophosphamide.Observed the general state,record the daily weight and calculate the rate of weight change before and after gavage.Thymus index and spleen index were measured by weighing immune organs.HE staining was used to detect the morphology of spleen tissue.The contents of IL-2,IFN-γ and TNF-α in spleen were determined by ELISA.The expression of IFN-γmRNA gene was detected by RT-PCR.Westernblot method was used to detect the expression of ATM,CHK1,Bcl2 and BAX proteins in spleen.Results: Compared with the congtrol group,the body weight,spleen index and thymus index of mice in the model group were significantly reduced.After the treatment with qi-gui blood-enriching particles,compared with the model group,the body weight was significantly increased,and the thymus index and spleen index were increased.Model group mice spleen organization form compared with the control group,lymph follicles decreased,lymphocyte generate had obstacle,RBC exudation was more,blood sinus widen,and there was seriouspassive congestion,lymphocyte nuclei necrosis,serious cell damage,interstitial fibers proliferation.Application of qi-gui blood-enriching particles improved above symptoms.Therefore,qi-gui blood-enriching particles can improve the spleen structure damage caused by cyclophosphamide.In the model group,the contents of TNF-α in the spleen were significantly increased compared with those in the control group,and decreased after the treatment with qi-gui blood-enriching granules,.On the congtrary,the contents of IFN-γ in the spleen appeared opposite change.Compared with the congtrol group,the relative expression of IFN-γ mRNA in the model group was significantly lower,but it growed significantly after the treatment with qi-gui blood-enriching granules.In mice treated with cyclophosphamide,the expression of ATM,CHK1 and BAX proteins in thespleen tissue was enhanced,and the expression of Bcl2 protein was weakened.After the intervention of qi-gui blood-enriching granules,the expression of ATM,CHK1 and BAX proteins in the spleen tissue was weakened,and the expression of Bcl2 protein was enhanced.Conclusion: qi-gui blood-enriching granules are beneficial to the development of spleen and thymus,can regulate the content of inflammatory factors,and improve the immunity of the body.It can adjust the expression of genes and proteins related to DNA damage repair.Therefore,qi-gui blood-enriching granules have a positive regulating effect on the immunosuppression caused by cyclophosphamide,which may be realized through the repair of DNA and the regulation of inflammatory factors. |
