| Objective:By cutting off the lateral collateral ligaments of the ankle in SD rats and culturing for 8 weeks,the osteoarthritis model of the ankle in SD rats will be established.To explore the expression of vitamin K dependent gamma glutamyl(GGCX)in SD rat model of ankle osteoarthritis,expect to provide experimental proof for the prevention and treatment of human ankle osteoarthritis.Methods:1.Ten healthy male SD rats,weighing about 200g,were selected They are about eight weeks old.By cutting off the lateral collateral ligament of the left ankle joint of SD rats,the animal model was established2.SD rats were classified into two groups:the control group(the right ankle joint,without treatment),the experimental group(the left ankle joint,the lateral collateral ligament of the left ankle joint was cut off)3.After 8 weeks,the articular cartilage of the control group and the experimental group were taken.Then,they were observed with naked eye and tested through pathological examination.The content of GGCXmRNA in the chondrocytes of the two groups were detected by PCR.4.Statistical differences of the content of GGCXmRNA in the two groups were assessed with the paired t-test,and P values<0.05 were considered statistically significant.Results:1.It was found by naked eyes that the articular cartilage surface of the experimental group was obviously rough and severely worn compared with that of the control group,and osteophytes were formed around it.2.Histological examination showed that the experimental group lost the homogenization of cartilage matrix,the arrangement of chondrocytes was disordered,the perichondrium of cartilage edge formed osteophyte,and the bone trabecula thickened3.PCR showed that GGCXmRNA was expressed in both groups But the experimental group was significantly lower than the control group,and the difference was statistically significant(P<0.05)Conclusion:1.By cutting off the lateral collateral ligament of the ankle in SD rats,the osteoarthritis model of the ankle in SD rats can be successfully made2.The content of GGCX in the articular cartilage of SD rats was significantly lower than that of the control group3.GGCX plays an important role in the pathogenesis of ankle osteoarthritis.The diagnosis of ankle osteoarthritis can be determined by the reduction of GGCX in articular cartilage. |
