Objective:Through the systematic and comprehensive study on the chemical composition of Artemisia ordosica petroleum ether and dichloromethane extract,the chemical information of petroleum ether and dichloromethane extract of Artemisia ordosica was found out to solve the problem of unclear chemical composition.At the same time,the HPLC quantitative analysis method of Artemisia ordosica was established to fill the blank of its content determination method.Methods:Extraction of Artemisia ordosica by refluxing with 95%ethanol,the 95%ethanol extract was obtained by petroleum ether in turn,dichloromethane,ethyl acetate and n-butanol were used to extract the solvent,then the solvent was recovered under reduced pressure to obtain different polarity extractants.Determination of Artemisia ordosicac from different sources by High performance liquid chromatography condition column;Topsil C18(250mm×4.6mm,5?m);The mobile phase consists of water(A)and acetonitrile(B);composition,with gradient elution order of 0-15 min,25%-38%B;15-40 min,38-45%B,The detection wavelength was 285 nm,the column temperature was 30?,current speed 1.0 mLˇmin-1.Results:A total of 30 compounds were isolated from the petroleum ether and chloroform extracts of Artemisia ordosica.Part Arteordoyn A(1)Dehydrofalcarindio(2)Dehydrofalcarinol(3)o-hydroxycapillene(4)Dehydromatricaria ester(5)Arteordoyn A(6)Arteordocoumarin A(7)6,7-dimethoxycoumarin(8)6,7,8-trimethoxycoumarin(9)6-hydroxy-7-methoxycoumari(10)4-hydroxylacetophenone(11)4-hydroxy-5-methoxylacetophenone(12)pathulenol(13)Uvaol(14)4-hydroxybenzaldehyde(15)4-hydroxy-5-methoxybenzaldehyde(16)4,5-dihydroxybenzaldehyde(17)5-hydroxy-7,4'-dimethoxyflavanone(18)dihydroconiferyl alcohol(19)5,4'-dihydroxy-7-methoxyflavanone(20)5,4'-dihydroxy-7,3'-dimethoxyflavanone(21)Coniferyl alcohol(24)5-hydroxy-7,4'-dimethoxyflavone(25)5,7-dihydroxy-4'-methoxyflavone(26)5,7,4'-trihydroxy-3'-methoxyflavone(27)5,7,4'-trihydroxyflavone(28)o-hydroxycinnamic acid(29)p-hydroxycinnamic acid(30).Hydroxycinnamic acid under selected experimental chromatographic conditions(1),o-hydroxy cinnamic acid(2),acacia(3),3,5-dihydroxy-7,4'-dimethoxyflavone(4),coniferol(5),5.4'-dihydroxy-7-methoxydihydroflavone(7),dihydroconiferol(8),5-hydroxy-7,4'-dimethoxydihydroflavone(9)hydroxy cinnamic acid(1),o-hydroxy cinnamic acid(2),acacia(3),3,5-dihydroxy-7,4'-dimethoxyflavone(4),coniferol'(5),5.4'-dihydroxy-7-methoxydihydroflavone(7),dihydroconiferol(8),5-hydroxy-7,4'-dimethoxydihydroflavone(9)were corrected in a wide concentration range.The regression equations are 1.y=1.14×103×-168 r=0.9995 2.y=4.11×102 x+127 r=0.9993 3.y=2.75×103 x-186 r=0.9998 4.y=5.07×102 x+127 r=0.9993 5.y=7.42×102 x-159 r=0.9997 6.y=2.88×103 x-131 r=0.9995 7.y=1.44×103 x-161 r=0.9995 8.y=9.22×102×-954 r-0.9996 9.y=1.78×103 x-254 r-0.9995.The average recoveries were 97.75%?96.00%?98.20%?97.00%?95.50%?99.33%?97.50%?6.50%?97.33%.RSD are less than 2.0%.Conclusion:Thirty compounds were isolated from different solvent extracts of Artemisia ordosica Krasch.Among them,compounds 1,5,7,23,24 are new compounds reported in literature at the end.Spathulenol and 5,4' Dihydroxy-7-methoxyl-dihydroflavones were used as the reference substance.Different unfolding systems were used to identify Artemisia ordosica from different habitats by TLC.Spathulenol and 5,4'Dihydroxy-7-methoxyl-dihydroflavones in petroleum ether-acetone(5:1),dichloromethane ethyl acetate formic acid(15:1:0.1).The spots are clear and reproducible when used as developing agent.
