Objective:To detect the expression of miRNAs-103a-3p and let-7f-5p in plasma of patients with prostate cancer,and to further analyze their relationship with clinicopathological parameters,and to explore the role and significance of these two miRNAs in the occurrence,progress,invasion and metastasis of prostate cancer.Methods:Preoperative plasma was selected from 66 patients with prostate cancer and 63 patients with benign prostatic hyperplasia(BPH)diagnosed by prostate biopsy from September 2017 to August 2018.The total RNA was separated and extracted from two groups of plasma samples,and the cDNA was formed by reverse transcription.The expression levels of miRNA103a-3p and let-7f-5p in plasma of-the two groups were detected by real-time quantitative PCR.The relationship between miRNA103a-3p and let-7f-5p and clinical and pathological parameters such as age,tumor stage,pathological grade,bone metastasis and PSA was analyzed.The study data was statistically analyzed using R 3.6.1 softwareResults:1.There was no significant difference in the expression of mirna103a-3p between the two groups(P=0.456),while the expression of let-7f-5p in prostate cancer group was significantly higher than that in BPH group(P=2.17e-07).2.The expression level of let-7f-5p in plasma of patients with prostate cancer was 3.27±2.33 in patients with age≤70 and 2.38±1.50 in patients with age>70.There was no significant difference between the two groups(P>0.05).The expression level of let-7f-5p in plasma of patients with Ⅰ and Ⅱ prostate cancer was 2.82±1.95,and the expression level of let-7f-5p in plasma of Ⅲ and Ⅳ stage was 2.70 ±2.02,the difference was not statistically significant(P>0.05).Gleason score>7 points the expression of let-7f-5p in plasma of prostate cancer patients was 2.32±1.81,and the expression level of patients with Gleason score<7 was 3.03±2.01,the difference was not statistically significant(P>0.05).The expression of let-7f-5p in plasma of two groups of prostate cancer patients with PSA<10ng/L and PSA>10ng/L was 2.56±1.23 and 2.83 ±2.08 respectively.There was no significant difference in the level of expression in the two groups(P>0.05).The expression of let-7f-5p in plasma of patients with bone metastases was 2.30±1.77,and the expression level in patients without bone metastasis was 2.97±2.08.There was no significant difference between the two groups(P>0.05).3.ROC curve analysis showed that AUC of let-7f-5p was 0.782(95%CI:0.703-0.861);AUC of PSA was 0.795(95%CI:0.720-0.871);AUC of let-7f-5p and PSA was 0.904(95%CI:0.852-0.957).Delong test analysis showed that the AUC of combined let-7f-5p and PSA was significantly higher than that of let-7f-5p(p=7.55E-04)or PSA(p=2.09E-03)alone.The decision curve analysis shows that the joint let-7f-5p and PSA models perform best in terms of risk prediction and clinical benefits.Conclusion:1.The expression level of miRNA-103a-3p in plasma is not related to the occurrence of prostate cancer.2.Let-7f-5p in the plasma of patients with prostate cancer was significantly up-regulated,but there was no significant correlation with the progression and metastasis of prostate cancer.ROC curve analysis shows that it has good diagnostic value,indicating that let-7 f-5p in plasma can be used as potential tumor biomarkers for early diagnosis of prostate cancer.
