Objective:The aim is to explore the value of multi-modal imaging and photothermal therapy in the acute phase of rheumatoid arthritis(stage of joint swelling)by using the cell probe delivery system(Gd@OVA-Cy@NEs)with neutrophil delivery Gd@OVA-Cy nanoprobe,and to achieve early diagnosis,course evaluation and accurate treatment of rheumatoid arthritis through individualized treatment to relieve joint swelling and relieve pai,.Methods:Firstly,Gd@OVA-Cy nanoprobe was synthesized by inorganic and aqueous method and characterized in vitro.Secondly,cytotoxicity and other experiments were performed to verify biocompatibility.We extracted bone marrow neutrophils from mouse and carried out nanoparticle loading experiments.After that,the rheumatoid arthritis mouse with different degrees of inflammation were divided into two groups(Gd@OVA-Cy group,Gd@OVA-Cy@NEs group)at random,and we injected probe intravenously and performed near-infrared fluorescence imaging,photoacoustic imaging and magnetic resonance imaging,then analyzed the imaging results.Finally,we performed photothermal treatment under the guidance of near-infrared light and closely observe the degree of swelling of the affected foot,and the effect was verified by pathologyResults:The Gd@OVA-Cy nanoprobe was constructed in this study and had good size and biocompatibility,and the neutrophil probe has a nanoparticle load rate of more than 96%.In vivo experiments have shown that the cell probe is better than single nanoprobe in active targeting performance and the long resident time of nanomaterials After being irradiated with near-infrared light,the photothermal effect of Gd@OVA-Cy@NEs was successfully performed to kill inflammatory cells,relieve the degree of joint swelling obviously,and show a significant anti-inflammatory effect Compared with the in vivo imaging,immunohistochemistry staining confirmed that FITC-labeled neutrophils have similar kinetics.Conclusion:This study demonstratesd that neutrophil-loaded Gd@OVA-Cy nanoprobes have achieved non-invasive,visualized imaging and integrated diagnosis and treatment of rheumatoid arthritis through neutrophil immune response.And it provided reliable data and theoretical support for evaluating the degree/course of inflammation in rheumatoid arthritis.In addition,the method of cell probe construction will promote its further research and application in the field of inflammation-related diseases.
