Objective:Colon cancer is the most common malignancy,second only to gastric and esophageal cancers,with more than 1.36 million new cases diagnosed worldwide each year.According to Chinese cancer data reports in recent years,the incidence and death rate of colon cancer in China are gradually increasing,and it has rapidly jumped to the top five malignant tumors.Patients with colon cancer,with few symptoms in the early stages and often diagnosed at an advanced stage,often miss the best time to treat,posing a major public health threat.EphB2,a new Eph subgene screened from the cDNA library of gastric cancer tissue,is expressed in a variety of cell components in embryonic and adult tissues,which are mainly involved in embryo,nerve development and vascular formation.Relevant data show that EphB2 is also involved in tumor formation.To investigate the expression of EphB2 protein in human colon cancer tissue,compare and analyze the expression of EphB2 protein in left and right colon cancer tissue,and analyze its prognostic value.Methods:From January 2008 to December 2013,180 cases of colon cancer specimens were selected for radical resection in Kunshan hospital affiliated to Jiangsu university.Among them,there were 83 male patients and 97 female patients,with a median age of 52 years,including 92 cases of LCC and 88 cases of RCC.Each sample was composed of colon cancer and corresponding adjacent normal tissues,and none of them received any radiotherapy or chemotherapy before surgery.Postoperative pathological results were confirmed by more than two pathologists.Complete clinicopathological data and prognostic follow-up data were available.Immunohistochemistry was used to detect the expression of EphB2 protein in colon cancer tissues and adjacent normal tissues,and the pathological parameters and prognosis were analyzed.All data were statistically analyzed by Graphpad 6.0 software and SPSS 20.0 software,and P<0.05 was considered statistically significant.Paired chi-square test was used to compare the expression of EphB2 protein in cancer tissues with clinical pathology such as age,tumor diameter,histological grade,lymph node metastasis and pathological classification.Survival of colorectal cancer patients was analyzed by kaplan-meier,Cox and log-rank methods.Results:(1)EphB2 protein was expressed in cytoplasm in both LCC and RCC tissues.Among them,the positive expression rate of EphB2 in LCC was 58.70%,and the positive expression rate of EphB2 in RCC was 67.05%.There was no difference of the positive expression rate of EphB2 between in the LCC and RCC tissues(P>0.05).The mean staining score of LCC was significantly lower than that of RCC and the paracancer normal tissues(P<0.001).In the LCC,EphB2 was negatively correlated with histological grade,lymph node metastasis and Dukes stage(P<0.05),but not with age,sex,tumor size and histological type(P>0.05).EphB2 protein expression was not related to age,sex,histological type,tumor size,histological grade,lymph node metastasis and Dukes stage in right colon cancer(P>0.05).(2)the 5-year survival rate of 92 cases of LCC was significantly higher than that of 88 cases of RCC(P=0.008).Although EphB2 expression was not related to 5-year overall survival(OS)in all patients or patients with RCC(P>0.05),The OS of the over-expressed EphB2 group was obviously higher than that in the low-expression group(P<0.05).The results of univariate and multivariate Cox survival analysis showed that histological grade,lymph node metastasis and Dukes stage of left and right colon cancer were independent influencing factors for overall survival.EphB2 expression level was not correlated with the OSof RCC.Low level of EphB2 in the LCC predicted poor prognosis.Conclusion:(1)In the LCC,EphB2 was negatively correlated with histological grade,lymph node metastasis and Dukes stage,while in the RCC,EphB2 was not correlated with clinical pathological parameters.(2)Low level of EphB2 in the LCC predicted poor prognosis,while in the RCC,there was no correlation between EphB2 expression and OS;(3)EphB2 played different roles in the development of left and right colon cancer,providing theoretical basis for the exploration of new targets in the diagnosis and treatment of colon cancer.
