Background:Safflower belongs to compositae,as a traditional Chinese medicine,has significant curative effects on brush-induced injuries and diseases of the blood circulation system.Pharmacological researches has proved that its water-soluble extract hydroxyl safflower yellow A has been clinically proved to have lipid-lowering,pressure-lowering and anti-tumor effects.Recently,there has team cooperation laboratory has been extracted a kind of flavonoids monomer compounds from safflower,an isomer of HSYA called hydroxy safflower yellow pigment B,besides antioxidant effect,less reports on the study of pharmacological effectsObjective:Our study aims to investigate the anti-cancer effect and molecular mechanis m of HSYB in breast cancer MCF-7 cells.Methods:MTT assay and colony formation assay were used to test the proliferation and survival of MCF-7 cells,respectively.Hoechst 33258 and flow cytometry were used to measure the cell apoptosis.Meanwhile,flow cytometry was used to determine cell cycle.Flow cytometry was also used to determine the mitochondrial membrane potential of MCF-7.Western blot was used to examine the protein levels.Results:The results showed,we found that treatment with HSYB remarkably reduced proliferation and survival of human breast cancer MCF-7 cells in a dose-dependent manner in vitro.Furthermore,we found that HSYB obviously arrested MCF-7 cell cycle at S phase,down-regulating cyclin D1,cyclin E and CDK2.Moreover,HSYB significantly suppressed the protein levels of p-PI3K,PI3K,AKT and p-AKT in HSYB-treated MCF-7 cells when compared with the control group.Apoptosis experiment results showed that hsyb-treated MCF-7 cells showed typical morphological characteristics of apoptosis in cell morphology.And flow cytometry showed that HSYB could reduce the mitochondrial membrane potential of MCF-7.In addition,we found that HSYB remarkably decreased the level of Bcl-2,increased the level of Bax,cleaved capase-3 and cleaved caspase-9,and subsequently induced MCF-7 cell apoptosis.Conclusion:HSYB can significantly inhibit the proliferation of human breast cancer cells MCF-7,and its mechanism may be related to the arrest of McF-7 cell cycle in S phase and the induction of apoptosis.
