| Background:The T lymphocytes play an important role in both the rejection of liver transplantation and viral hepatitis. The present study show that the lymphocyte is a big family and of each subset has different functions.Regulatory T cells(Treg)are a large group of T cells that produce inhibitory cytokines, and the role of each Treg subset is different.Treg not only expresses cluster of differentiation 25(CD25)and forkhead transcription factor p3(Foxp3),but also some Treg expresses the glucocorticoid-induced tumor necrosis factor receptor(GITR)or programmed cell death 1(PD-1),in which,a subset of Treg with the three types of surface markers were highly expressed Treg was called TriplehiTreg. The recent research suggests that TriplehiTreg(CD25hi GITRhiPD-1hi)is a powerful Treg cell population, which can highly self-reactive and controll lympho-proliferation in peripheral lymph nodes, contrarily, eliminating TriplehiTreg could induce liver infiltrated by lymphocytes, and cause severe inflammation reaction. The study of the TriplehiTreg subset in liver diseases has great significance for the treatment and control of liver diseases. Objective:This study intends to explore the relationship between lymphocyte subgroup and Treg subgroup with rejection and inflammatory responses in liver diseases by analyzing the changes of peripheral blood lymphocytes and their subgroup in clinical liver transplant recipients and patients with viral hepatitis type B in our hospital,to provide theoretical basis for their monitoring and treatment. Methods:The clinical data of patients with liver transplantation and chronic hepatitis B in the Henan university people’s hospital(Henan provincial people’s hospital)from January 2019 to January 2020 were analyzed, including the proportion of B cells,T cells,NK cells and CD4+T cells and CD8+T cells subset,as well as the proportion of Treg cells and its subset with high expression of GITR,PD-1 and CD25 in peripheral blood lymphocytes by using flow cytometry instrument, compared with the healthy control group,to explore its cell percentage changes. The data were expressed as mean±standard error(x±SE),and the data were accurate to two decimal places. The statistical analysis was carried out by software SPSS22.0Results:TriplehiTreg cells can inhibit the proliferation of toxic T cells in the liver transplantation recipient, induce and maintain immune tolerance, activate the proliferation during the immune clearance period of Hepatitis B,control the excessive inflammatory response, and alleviate the pathological reaction.Further research is needed to determine whether TriplehiTreg cells play a role in the inflammatory response to obstructive jaundice. |
