Objective Breast cancer is the most common malignant tumor in women,and molecular subtype is one of the main basis for clinical diagnosis and treatment of breast cancer at present.Local recurrence or distant metastasis is the main death factors of female breast cancer.However,the mechanism of recurrence and metastasis of breast cancer has not been clarified.AKAP12 is located in 6q24-25.2,which is the lost hotspot of many types of cancer.In recent years,it has been reported that the expression of AKAP12 is downregulated in lung cancer,prostate cancer and basal cell carcinoma.AKAP12 may be a potential tumor suppressor gene,but there are few reports of AKAP12 and breast cancer.In this study,we analyzed the expression of AKAP12 mRNA and protein in different molecular subtypes of breast cancer,the relationship between AKAP12 protein and clinicopathological characteristics and prognosis,the relationship between AKAP12 protein expression in breast cancer metastasis and clinicopathological characteristics by using bioinformatics website analysis and immunohistochemistry analysis,in order to provide basis for clinical diagnosis and treatment.Methods UALCAN database was used to analyze the expression of AKAP12 mRNA and protein in breast cancer tissues and normal tissues,Bc-GenExMiner v4.4 database was used to analyze the expression of AKAP12 mRNA in four molecular subtype breast cancers.All specimens were collected from January 2013 to December 2018 whom diagnosed in Shanghai tenth people’s hospital.Of which 560 breast cancer and 17 metastasis tissues.Clinical and pathological data of all breast cancer patients,immunohistochemical was collected to analyze the expression of AKAP12 protein in four molecular subtypes of breast cancer tissues and adjacent tissues.Chi-square analysis of AKAP12 protein expression in different molecular types of breast cancer and clinicopathological factors.Kplan-meier was used to analyze the relationship between AKAP12 protein and prognosis of different molecular types of breast cancer.Results UALCAN database explored that the expression of AKAP12mRNA was significantly lower than that in para-cancerous tissues(p<0.05),and the expression of AKAP12mRNA in four molecular subtypes of breast cancer were significantly lower than that in para-cancerous tissues(p<0.05).Bc-GenEcMiner v4.4 database showed that AKAP12mRNA was expressed differentially in molecular subtypes of breast cancer.AKAP12mRNA showed a decreasing trend in Luminal A,HER2-positive and triple-negative breast cancer.Further analysis showed that compared with para-cancerous tissues,the expression of AKAP12 protein in all four molecular subtypes of breast cancer tissues was significantly lower.And the high expression rate of AKAP12 protein in 17 metastatic lesions was 29.4%(5/17),which was significantly lower than that in adjacent tissues(p<0.05).The expression of AKAP12 was correlated with tumor diameter,lymph node metastasis,TNM stage,histological grade,vascular invasion and molecular subtype.However,there was no significant difference with age,menstrual status,ER,PR,HER-2,KI67,chemotherapy or radiotherapy.Further analysis of correlation between clinicopathological factors showed that the expression of AKAP12 protein was related to the histological grade in Luminal A.In Luminal B,AKAP12 protein expression was correlated with tumor size,lymph node metastasis,TNM stage and radiotherapy(p<0.05).In HER2-positive breast cancer,it was found that AKAP12 protein was related to tumor size and TNM stage.The expression of AKAP12 protein in triple negative breast cancer was correlated with lymph node metastasis(p<0.05).The high expression rate of AKAP12 protein was 29.4%(5/17).There was no significant difference between AKAP12 protein and age,menstrual status,tumor size,lymph node metastasis and TNM stage(p>0.05).Kplan-meier survival analysis showed that there was no significant difference between AKAP12 expression of AKAP12 and disease-free survival.However,in HER2-positive breast cancer,the prognosis of patients with low expression of AKAP12 protein was worse than those with high expression of AKAP12 protein(p<0.05).Conclusion1.The expression of AKAP12 mRNA was down-regulated in breast cancer,and there were different expression in different molecular types of breast cancer.2.Compared with the para-cancerous tissues,the expression of AKAP12 protein in the four molecular types of breast cancer is low.Low expression of AKAP12 protein also has high invasive clinical characteristics.3.The abnormal expression of AKAP12 may be related to the development of various molecular subtypes of breast cancer,and in HER2 positive breast cancer,low expression of AKAP12 protein is related to poor prognosis.Figure 16 table 11 reference 53...
