In this study,six flavonoids from Flos Sophorae Immaturus were extracted efficiently by using the new green extract medium-deep eutectic solvent.The extraction process of microwave assisted deep eutectic solvent was developed and its technological conditions were optimized,the flavonoids in the extract were isolated and enriched,and the antioxidant activity of the enrichment was also investigated.An accurate,rapid and efficient method for the detection and analysis of major flavonoids in Flos Sophorae Immaturus was established.It provides important theoretical guidance for efficient development and rational utilization of Flos Sophorae Immaturus.The results are as follows:1.A method for the detection and analysis of Rutin,Nicotiflorin,Narcissin,Quercetin,Kaempferol and Isorhamnetin in Flos Sophorae Immaturus was developed.Chromatographic column:Diamonsil C18(250×4.6 mm i.d..5 μm),0.1%aqueous phosphate solution(A)and acetonitrile(B)as mobile phase,The columemperature is 30℃,the injection volume is 5μl and the flow rate is 0.6 ml/min.The detection wavelength of the target component is 360 nm.The elution conditions are as follows:0-15 min,15-20%(B);15-25 min,20-35%(B);25-30 min,35%(B);30-45 min,35-50%(B);45-50 min,50-15%(B).2.The extraction process of Rutin,Nicotiflorin,Narcissin,Quercetin,Kaempferol and Isorhamnetin from microwave-assisted deep eutectic solvents was developed and optimized.The optimum technological conditions were determined.Deep eutectic solvent:choline chloride/1,4-butanol=1/2(mol/mol,water content 25%)Time:20 minTemperature:62℃Liquid-solid ratio:26ml/gUnder the optimum parameters,six flavonoids were obtained,Rutin,Nicotiflorin,Narcissin,Quercetin,Kaempferol and Isorhamnetin and the average extraction yields were 116.78 mg/g,15.01 mg/g,23.85 mg/g,27.59 mg/g,3.09 mg/g and 3.33 mg/g,the extraction results exceeded the deep eutectic solvent thermal reflux and deep eutectic solvent ultrasonic extraction.3.A macroporous adsorption resin was used to separate and enrich the Flos Sophorae Immaturus extract and to compare the antioxidant activity of the concentrate.The separation and enrichment of flavonoids from Flos Sophorae Immaturus by resins such as D101,AB-8,NKA-9,D-3520,DM-130 and HPD-826 were investigated.D101-type resins for Rutin,Nicotine,Narcissin,Quercetin,Kaempferol and Isorhamnetin had the highest recovery of the six components,87.71%,81.24%,85.87%,76.45%,81.23%,79.39%,respectively.Meanwhile,the antioxidant activity of the enrichment of the D101 macroporous resin was stronger than that of other resins.In this paper,microwave assisted extraction of low eutectic solvent was carried out to optimize the extraction process of main flavonoids in Flos Sophorae Immaturus,Meanwhile the extract was isolated and enriched,and the antioxidant activity of the extract was investigated.This study provided scientific basis and theoretical guidance for the effective development and utilization of Flos Sophorae Immaturus.
