| Objectives:The quality of corneal endothelial cells from clinical donors was monitored.To analyze the influencing factors of corneal endothelial cell density and explore its change rule.Methods:From January 2019 to November 2019,a total of 76 corneal grafts(donor number 253-291(38 cases)were obtained from the First People’s Hospital of Kunming.Inclusive criteria:corneal donor obtained from the First People’s Hospital of Kunming;donor age between 3 and 65 years;No corneal Central District Injury;intact anteroposterior elastic layer;transparent stroma.Criteria excluded:The donor had a history of bacterial,Fungal,and viral infection;the donor had a history of endothelial injury related surgery or high intraocular pressure;and the donor had a history of ocular tumor or neurological disorder.Two were excluded due to extensive corneal ulcers,four were excluded due to infectious disease,one was excluded due to prior internal eye surgery,and one was excluded due to prior Glaucoma history.The remaining 68 corneal donors were included in the study.The corneal tissue acquisition registration form was developed to record the donor number,gender,age,cause of death,time of death,donor preservation time,and endothelial cell density detection.The donor corneal grafts were preserved in 4℃ EUSOL-C medium-term preservation solution for 0-1 day,1-3 days,3-7 days,more than 7 days,and the corneal endothelial cell density was measured on the same day,and carries on the statistical analysis to the measured data.Results:1、Among the 1,68 corneal donors included in the study,27 were male and 7 were female.According to age group,the average age is 36.09 13.5 years old between 5 and 64 years old,including 28 cases in<30 years old group(3253.964 340.288 ce ⅡS/mm2),36 cases in 30-60 years old group(2965.321282.685 ce IIS/mm2),and 4 cases in>60 years old group(2279.750 157.199 ce ⅡS/mm2).the density difference of donor corneal endothelial cells in different age groups is statistically significant,p<0.05.According to the death-acquisition time,24 cases(3277.25 343.532 ce ⅡS/mm2)were obtained in the 0-2 hour group,28 cases(3008.61 340.506 ce ⅡS/mm2)were obtained in the 2-4 hour group,and 16 cases(2736.69 290.336 ce ⅡS/mm2)were obtained in the 4-6 hour group.the difference of corneal endothelial cell density in different death-acquisition time groups was statistically significant,p<0.05.According to the causes of death,24 cases were divided into three groups:donor death causes trauma(3025.06 420.150 ce IIS/mm2),10 cases were cardiovascular and cerebrovascular accidents(3073.95 292.831 ce ⅡS/mm2).there was no significant difference in the density of donor corneal endothelial cells with different causes of death,p>0.05.2.Using multiple linear regression analysis,we set corneal endothelial cell density as dependent variable Y,independent variables as age X1 and donor death-acquisition time X2.Regression equation:Y=3849.128-11.711X1-143.474X2,fitting degree R2=0.5,regression model has statistical significance,p<0.05,the change rule of corneal endothelial cell and donor death-acquisition time conforms to linear rule.The density of donor corneal endothelial cells is negatively correlated with donor age and donor death-acquisition time.The death-acquisition time of 3-65 year old donors is between 7.5-12.6h,and the density of endothelial cells of the obtained graft can be>2000mm2.3.in eusol-c preservation solution at 4℃,the preservation time is between 0-14 days,with an average of 6.13 days.23 cases were preserved for 1-3 days,32 cases for 3-7 days,and the remaining 11 cases were preserved for 7-14 days.The measured corneal endothelial cell density was 68 cases(3039.44 385.554 CE IIS/mm2)on the day of material collection,66 cases(2873.969 354.876 CEⅡS/mm2)for 1-3 days,43 cases(2625.600 318.574 CE ⅡS/mm2)for 3-7 days,and 11 cases(2455.727 290.285 CE ⅡS/mm2)for 7-14 days.The average corneal endothelial cell density before preservation was 3039.44 385.554 ce IIS/mm2 and 2723.57 343.535 ce ⅡS/mm2 before use.the density of corneal endothelial cells after preservation was significantly lower than that before preservation,and the difference was statistically significant(p<0.05),but the change rule of donor corneal endothelial cells with preservation time did not conform to the linear trend.Conclusions:1、Age and death-donor preservation time could affect the density of donor corneal endothelial cells,but the cause of donor death was not related to the density of donor corneal endothelial cells2、The changes of age and death-donor preservation time were in line with the linear trend,the density of donor corneal endothelium cells was negatively correlated with the death and preservation time of the donor,and the older the donor was,the shorter the preservation time of the donor was required3、In 4℃ EUSOL-C preservation solution,the density of donor corneal endothelial cells decreased with the increase of preservation time,but the change law did not accord with the linear trend. |
