Objective:By detecting the expression level of sCLU in pancreatic cancer,and analyzing the relationship between s CLU expression and clinicopathological features and prognosis of patients with pancreatic cancer,it provides a new basis for early diagnosis and prognosis judgment of patients with pancreatic cancer.Methods:1.The expression levels of sCLU in pancreatic cancer tissues and adjacent tissues of 5 patients and 3 pancreatic cancer cell lines and normal pancreatic cell lines were detected by Western Blot.2.Paraffin sections of 50 pancreatic cancer tissues and corresponding adjacent tissues were detected by immunohistochemical staining The expression of sCLU and to analyze the relationship between the differential expression and the clinicopathological characteristics and prognosis of patients with pancreatic cancer.Results:1.The results of Western Blot and immunohistochemical analysis at both cell and tissue levels showed that sCLU was overexpressed in pancreatic cancer.Western Blot results showed that the expression of s CLU in 3 pancreatic cancer cell lines was significantly higher than that in normal pancreatic cell lines,and the expression of sCLU in 5 pairs offresh tissue samples in 80%(4/5)of pancreatic cancer tissues was significantly higher than that in the corresponding para-carcinoma tissues(P<0.05).Immunohistochemistry of 50 pairs of paraffin sections showed that sCLU was positively expressed in 34 cases(68%)of the pancreas and 8 cases(16%)of the paracancer,with statistically significant differences(P<0.001).2.Analysis of sCLU expression and clinicopathological parameters in pancreatic cancer showed that the expression level of sCLU was significantly related to tumor size,tumor differentiation,and vascular and nerve invasion of pancreatic cancer(P <0.05),but not with gender,age,CA19-9 level,clinical stage,lymph node metastasis(P>0.05).3.Survival analysis showed that the 1-year,2-year,3-year overall survival rate of patients with sCLU positive expression was significantly lower than that of patients with sCLU negative expression(61.8%,34.2%,20.5% vs.81.3%,62.7%,33.4%),and the median survival time was significantly shorter than that of patients with s CLU negative expression(18 months vs 27 months),(P<0.05).Conclusion:1.sCLU was overexpressed in pancreatic cancer cells and tissues.2.The expression of s CLU is significantly related to tumor size,differentiation and vascular and nerve invasion in pancreatic cancer.3.The expression of sCLU is negatively correlated with theprognosis of pancreatic cancer.The survival time of patients with positive expression is shorter than that of patients with negative expression.
