Purposes: The differentially expressed proteins in nucleus pulposus cells in degenerative lumbar intervertebral disc and normal lumbar intervertebral disc were screened.To establish the information database of differentially expressed proteins in nucleus pulposus cells of the degeneration lumbar intervertebral disc.Metheods : In this study,samples of nucleus pulposus cells from degenerative lumbar intervertebral disc and normal lumbar intervertebral disc(control group)were collected.The TMT technique of proteomics was used to construct the map of differentially expressed proteome of nucleus pulposus cells between the lumbar intervertebral disc degeneration group and the control group.The contrasting proteins of down-regulated and up-regulated expression and all the contrasting proteins were found by screening.The differentially expressed proteins were marked with mass spectrometry.Then the differential proteins of biological processes,molecular functions and cellular components were found by bioinformatics GO annotation.GO functional enrichment and KEGG pathway enrichment were utilized to screen the main pathways involved in target proteins.Finally,the differentially expressed proteins shared by GO function and KEGG pathway enrichment were accurately found.Results:In this study,TMT technology of proteomics was used.It was found that the total number of secondary mass spectra identified in the nucleus pulposus cells of the lumbar intervertebral disc was 284386.The total number of identified peptides was 12678,and the total number of unique peptides that could be identified was11626.The total number of mass spectra that can be matched by the identified peptides is 51303 and the total number of proteins that can be identified is 1885.Through protein quantitative analysis.It was found that in the nucleus pulposus cells of the lumbar intervertebral disc,there were 73 differentially expressed proteins between non-degenerative control group B and degenerative A group,including 9up-regulated differentially expressed proteins and 64 down-regulated differentially expressed proteins.GO annotations show that all differential proteins are involved in biological regulation in biological processes.In terms of molecular function,all differential proteins are associated with binding activity.In terms of cellularcomposition,all differential proteins are involved in the composition of organelles and cellular parts.Through the accurate Fisher test,among 73 contrasting proteins in nucleus pulposus cells,48 contrasting proteins were significantly enriched in 30 GO terms belonging to biological processes.Using KEGG pathway enrichment,three pathways were identified: autophagy-other pathways,primary bile acid biosynthesis and neuroactive ligand-receptor interaction.There are 5 kinds of differentially expressed proteins in these three pathways.Conclusions : In this study,proteomic analysis of nucleus pulposus cells in degenerative lumbar intervertebral disc and normal lumbar intervertebral disc was carried out.64 contrasting proteins were down-regulated.There were 9 contrasting proteins up-regulated.The total number of contrasting proteins was 73.Five differentially expressed proteins were identified in the three pathways.The information base of differentially expressed proteins in nucleus pulposus cells of the degenerative lumbar intervertebral disc was established. |
